Cortisol and stress modulation of fear memory reconsolidation
- Cortisol, a stress hormone, is a potent modulator of learning and memory. However, its possible influences on memory reconsolidation, a post-retrieval process, remain unclear. Using a three-day reconsolidation design, three studies investigated the effects of cortisol and stress on fear memory reconsolidation in humans. Fear memories were created using the fear conditioning paradigm, retrieved following a pharmacological or behavioral manipulation, extinguished and tested for reinstatement. We demonstrated an enhancing effect of cortisol and an impairing effect of stress on fear memory reconsolidation in men, but no reconsolidation effect in women. Cortisol concentrations, sympathetic nervous system activity, affective and cognitive factors may account for the difference in the direction of the effect. The lack of effect in women could be explained by their mixed hormonal status. The presented findings contribute to the understanding and treatment of aversive memories.
- Das Stresshormon Cortisol moduliert Lern- und Gedächtnisprozesse. Sein möglicher Einfluss auf die nach dem Abruf stattfindende Gedächtnisrekonsolidierung ist jedoch unklar. Drei Humanstudien untersuchten die Effekte von Cortisol und Stress auf die Furchtgedächtnisrekonsolidierung. Mittels Furchtkonditionierung wurde an drei Tagen ein Furchtgedächtnis aufgebaut, nach pharmakologischer oder behavioraler Manipulation abgerufen, extinguiert und auf Reinstatement getestet. Es zeigte sich ein verbessernder Effekt von Cortisol und ein beeinträchtigender Effekt von Stress auf die Furchtgedächtnisrekonsolidierung bei Männern, aber kein Rekonsolidierungseffekt bei Frauen. Cortisolkonzentration, Aktivität des sympathischen Nervensystems sowie affektive und kognitive Faktoren könnten die Dissoziation des Effekts erklären. Der fehlende Effekt bei Frauen könnte dem unterschiedlichen Hormonstatus zugeschrieben werden. Diese Ergebnisse tragen zu Verständnis und Behandlung aversiver Erinnerungen bei.
