Wachendorfia thyrsiflora (Haemodoraceae) enthält eine Gruppe von sekundären Naturstoffen, sogenannte Phenylphenalenone, die vermutlich an der Abwehr von Pathogenen beteiligt sind. An den ersten Schritten der Biosynthese dieser Substanzen, der Bildung von linearen Diarylheptanoiden, sollten sogenannte Typ III Polyketidsynthasen beteiligt sein. Die vorgelegte Arbeit beschäftigt sich mit der Isolation von Polyketidsynthasen aus Wurzelkulturen von Wachendorfia thyrsiflora und deren Charakterisierung. Es wurden drei Enzyme analysiert und im Hinblick auf ihre Beteiligung an der Biosynthese untersucht. Eines der drei Enzyme ist vermutlich an der Biosynthese beteiligt, jedoch allein nicht in der Lage Diarylheptanoide zu bilden. Die Arbeit enthält neben der Charakterisierung dieser Proteine durch Enzymassays auch Hinweise auf die Natur eines weiteren benötigten Enzyms. Wachendorfia thyrsiflora (Haemodoraceae) contains a group of secondary metabolites called phenyl phenalenons, that are probably involved in the defense against pathogens. Type III polyketide synthases should be involved in the first steps of the biosynthesis, the formation of linear diarylheptanoids. This work deals with the isolation of polyketide synthases from root cultures of Wachendorfia thyrsiflora and their characterization. Three enzymes were analyzed and investigated with regard to their involvement in the biosynthesis. One of the enzymes is probably involved in the biosynthesis, but itself not able to form diarylheptanoids. This work contains in addition to the characterization of the proteins by enzyme assays also hints to the nature of a second involved enzyme.
