Die Alzheimer Demenz (AD) ist eine der am häufigsten auftretenden Demenzen mit in Zukunft laut Prognosen steigenden Fallzahlen, was erhebliche soziale und ökonomische Auswirkungen für die Betroffenen und auch die Gesellschaft haben wird. Es ist bis jetzt keine ursächliche Therapie für die Patienten vorhanden, ebenso ist eine sichere Diagnose schwierig. Das liegt zum einen an den noch nicht vollständig aufgeklärten Ursachen der Krankheit, zum anderen daran, dass eine sichere spezifische Diagnose erst post mortem durch histologische Untersuchungen am Gehirn erfolgen kann. Die derzeit anerkannten Hypothesen gehen davon aus, dass die bei den Patienten gefundenen Abeta- und Tau-Protein-Ablagerungen in ursächlichem Zusammenhang mit der Krankheit stehen. Diese entstehen durch Fehlprozessierung des beta-Amyloid-Vorläuferproteins sowie durch Fehlfunktionen im Gleichgewicht von Phosphatasen und Kinasen. Davon ausgehend kann sowohl ein therapeutischer als auch ein diagnostischer Ansatz entwickelt werden. Im Hinblick auf einen therapeutischen Ansatz wurden im Rahmen dieser Arbeit gamma-Sekretase Modulatoren synthetisiert. Dabei standen speziell Carbonsäurederivate und medizinalchemisch isostere Strukturmotive im Vordergrund. Speziell für die wichtige Isosterenklasse der Tetrazole wurde eine neuartige, sichere und effiziente Synthesemethode in ionischen Flüssigkeiten entwickelt. Im Hinblick auf die Diagnostik sind die derzeit gängigen Untersuchungsmethoden, basierend auf kognitiven, psychologischen Tests und bildgebenden Verfahren wie Computer Tomografie (CT), Magnet Resonanz Tomografie (MRT), Positronen-Emissions-Tomografie (PET) und Einzelphotonen-Emissions-Tomografie (SPECT), nicht hinreichend. Da sowohl die Befunde mit den verwendeten verfügbaren Markern nur bedingt mit dem Schweregrad der Demenz korrelieren als auch im Hinblick auf eine Frühdiagnostik sind diese Verfahren zu kostenintensiv. Davon ausgehend wurde im Rahmen dieser Arbeit an neuen Markern und der Entwicklung von fluoreszenzbasierten diagnostischen Methoden gearbeitet. Dabei konnte eine Reihe von in vitro aktiven Fluoreszenzmarkern auf der Basis von benzoid- und pyrimidinbasierten Bisstilbenen synthetisiert werden. Zugehörige Untersuchungen hinsichtlich ihrer pharmakologischen Eigenschaften wurden sowohl histologisch evaluiert als auch mit Hilfe von Zebrafischeiern und entsprechend genetisch modifizierten Maustiermodellen untersucht. Es konnten mit Hilfe der Farbstoffe neue Erkenntnisse bezüglich des Plaquewachstums in kranial gefensterten Tg2575 Mäusen gewonnen werden sowie eine verbesserte Methode für wiederholte Bildaufnahmen in diesen Tiermodellen etabliert werden. Es konnte dadurch ebenfalls gezeigt werden, dass diese neuen Strukturmotive für eine Entwicklung eines fluoreszenzbasierten Verfahrens der Diagnose in vivo am Auge und oder der Nase geeignet sind.

Alzheimer's disease (AD) is one of the most common forms of dementia and is forecast to increasing case numbers, which will have significant social and economic consequences for those affected and for society. There is as yet no causal therapy for the patients existing, and a reliable diagnosis is difficult. This is partly due to the still not fully understood causes of the disease and partly because a reliable specific diagnosis is only possible by post mortem histological studies. The currently accepted hypotheses assume that the Abeta and tau protein deposits found in the patients are causally associated with the disease. These deposits are caused by faulty processing of the beta-amyloid precursor protein as well as malfunctions in the balance of phosphatases and kinases. On this basis both a therapeutic and a diagnostic approach are the aim to be explored. With respect to a therapeutic approach gamma-secretase modulators were synthesized. In this work especially carboxylic acid derivatives and isosteric structural motifs were in focus of investigation. Especially for the important class of tetrazole isosteres a novel, safe and efficient synthetic method in ionic liquids was developed. With regard to the diagnosis the currently used methods based on cognitive, psychological testing and imaging techniques such as computer tomography (CT), magnetic resonance imaging (MRI), positron emission tomography (PET) and single photon emission computed tomography (SPECT) are not sufficient, because the available markers which are used at present only partially correlates with the severity of dementia and the marked regions and with respect to early diagnosis this process is too costly. Based on that in this work new markers and the development of fluorescence-based diagnostic methods were investigated. Thereby a number of in vitro active fluorescent markers on the basis of benzenoid and pyrimidin-bisstilbenes were synthesized. Related investigations with regard to their pharmacological properties were examined by histological evaluations as well as by using zebra fish eggs and accordingly genetically modified mouse animal models. New insights by using the dyes for investigating the plaque growth were obtained in cranial windowed Tg2575 mice and an improved method for repeated imaging in these animal models could be established. It could thus be also shown that these new structural motifs are suitable for the development of a fluorescence-based method of diagnosis in vivo in the eye or nose.
