Einfluß von Apolipoprotein H auf die Endothelmodifikation und Endothelaktivität

  • ApoH ist ein endogenes Glykoprotein mit einem Molekulargewicht von 54 kDa. Es wird sowohl in der Leber als auch in der Plazenta synthetisiert. Das monomere Protein besitzt 4 sich wiederholende Domänen und eine 5. Domäne, an die sich ein C-terminaler Schwanz anschließt. Dieser 20 Aminosäuren lange Schwanz kann durch Plasmin enzymatisch zu einer geclippten Form (cApoH) umgesetzt werden. Die physiologische Funktion des Proteins ist bis heute nicht eindeutig geklärt. Vorausgegangene Untersuchungen demonstrierten jedoch, dass ApoH Einfluß auf die Angiogenese und somit Proliferation vaskulärer Endothelzellen ausübt. Um den Einfluß von ApoH und cApoH auf Endothelzellen untersuchen zu können, wurden im Rahmen der vorliegenden Arbeit Proliferationsversuche im Zellkulturmodell mit humanen Endothelzellen und bovinen Kapillarendothelzellen durchgeführt. Des weiteren wurden Auswirkungen von ApoH und cApoH auf einzelne Zellzyklusphasen der Endothelzellen untersucht. Um auch die Auswirkungen von ApoH und cApoH innerhalb der Zelle beurteilen zu können, wurde in darauffolgenden Experimenten das Augenmerk auf die intrazelluläre Signalweiterleitung gelegt. Dabei wurden insbesondere die MAP-Kinasen p 38, JNK, ERK 1/2 untersucht, da es einen Zusammenhang zwischen Apoptose und ihrer Aktivierung gibt. Der intrazelluläre Proteingehalt wurde mittels Western Blot sowie Flow-Cytometrie gemessen. In den Proliferationsversuchen konnte eine deutliche Wachstumshemmung der Endothelzellen durch ApoH und cApoH festgestellt werden. Bei der Zuordnung der Zellen zu den einzelnen Zellzyklusphasen fiel auf, dass es durch ApoH und cApoH zu einer Verschiebung der Phasen zugunsten der G 2-, M- und S-Phasen kam. Eine 90-minütige Inkubation der Endothelzellen mit ApoH und cApoH bewirkte bei JNK und ERK 1/2 eine deutliche Erhöhung der Aktivität in der Flow-Cytometrie-Untersuchung. Im Falle von p 38 kam es durch ApoH jedoch zu einer Verminderung der Aktivität. Western Blot Experimente ergaben bei allen drei MAP-Kinasen eine vermehrte Expression im Vergleich zu den unbehandelten Kontrollen bzw. mit Plasmin behandelten Zellen. Im Rahmen der Arbeit wurde nachgewiesen, dass es unter dem Einfluß von ApoH und cApoH zu einer Hemmung der Angiogenese von Endothelzellen kommt. Es konnten sowohl Auswirkungen auf die Endothelzellmodifikation als auch auf die Endothelzellaktivität belegt werden.
  • ApoH is an endogenous glycoprotein with a molecular weight of 54 kDa. It is synthesised in the liver and in the placenta. Beside 4 recurring domains, the monomere protein ApoH owns a 5th domain to which a C-terminal tail is adopted. Therefore, the whole tail consists of 20 amino acids. By the use of plasmin, it can be transformed enzymatically to a clipped form (cApoH). To date, the physiological function of the protein is not clarified in a definite way. However, earlier research shows that ApoH influences the angiogenesis and that it bears on the proliferation of vasculary endothelial cells. The present work, though, places great emphasis on the influence of ApoH and cApoH on endothelial cells. Therefore, proliferation tests in the cell culture model were carried out with both, human endothelial cells and bovine capillary endothelial cells. One can also find an analysis of the effects of ApoH and cApoH on all cell cycle phases of endothelial cells in this work. Furthermore, different tests served to pay special attention to the intracellular signal forwarding, so that implications of ApoH and cApoH could be described, too. In doing so, attention was directed to the MAP-kinases p 38, JNK, ERK 1/2 as there is a connection between apoptosis and their activation. It should be mentioned that the amount of intracellulary protein was measured by the use of western blot and flow-cytometry. The proliferation test showed that the use of ApoH and cApoH led to a clear growth inhibition of endothelial cells. By the time the cells were allocated to all cell phases, it was appealing that some cell phases were changed. It can therefore be concluded that ApoH and cApoH help to detect more endothelial cells in G 2-, M. and S-phases. In the flow-cytometry investigation, a 90-minute incubation of the endothelial cells with ApoH and cApoH revealed a clear increase of activity with JNK and ERK 1/2. However, it should be remarked that ApoH caused a decrease of activity with p 38. Western blot tests showed an increased expression with all 3 MAP-kinases when compared to untreated controls and to cells treated with plasmin. One can sum up that tests done for this work emphasise the fact that the angiogenesis of endothelial cells is inhibited when in contact with ApoH and cApoH. In this matter, not only effects on the modification of endothelial cells have been proven but also effects on the activity of endothelial cells.
