Kurzfassung
In vorliegender Arbeit werden mittels der BioLector-Technologie gängige Hochdurchsatz-Kultivierungsprozesse in Mikrotiterplatten untersucht. Es wird gezeigt, dass umfassendes Monitoring von Schlüsselparametern in Mikromaßstab-Kulturen - wie Biomasse und Produktbildung - ein profundes Verständnis und damit Optimieren von rekombinanten Expressionssystemen und Hochdurchsatz-Kultivierungsprozessen ermöglicht. Es werden neue Konzepte erarbeitet und getestet, um mit Problemen verschiedener Wachstumskinetiken von Kulturen in Mikrotiterplatten besser umzugehen. Darüber hinaus wird eine neuartige Plattform für die Hochdurchsatz-Versuchsdurchführung von Mikromaßstab-Kulturen etabliert, indem ein Pipettierroboter mit einem BioLector kombiniert wird (sog. Robo-Lector-Plattform). Letztendlich kann der Robo-Lector zum angestrebten Paradigmen-Wechsel in der Bioprozessentwicklung beitragen. Dies schließt den Wechsel ein von eher empirischer Prozessentwicklung (niedriger Durchsatz mit keinem oder unausgereiftem Monitoring) hin zur Hochdurchsatz-Versuchsdurchführung mit sehr ausgeklügeltem, aber einfachem Monitoring, wobei dadurch tiefere Erkenntnisse über biologische Produktionssysteme resultieren.In this work, common high-throughput cultivation processes in microtiter plates are investigated with the BioLector technology. It is demonstrated that extensive monitoring of key parameters (i.e. biomass and product formation) in microscale cultivations enables a profound understanding and hence optimization of recombinant expression systems and high-throughput cultivation processes. New concepts to deal with issues of different growth kinetics of cultures in microtiter plates are elaborated and tested. Furthermore, a novel high-throughput experimentation platform for microscale cultivations is established by combining a pipetting robot with the BioLector, resulting in the Robo-Lector platform. Ultimately, the Robo-Lector can contribute to the envisioned paradigm shift in bioprocess development. This entails switching from rather empirical process development (low-throughput experimentation with no or unsophisticated monitoring) to high-throughput experimentation with very sophisticated yet simple monitoring to gain deeper knowledge of biological production systems.
