Die HIV-Pandemie : Ein Problem weltweit, auch in Deutschland

Infektionen mit dem humanen Immundefizienzvirus (HIV) sind nach wie vor ein globales Problem und ein Impfstoff lässt bisher noch auf sich warten. Da es bis heute noch keine breit anwendbare Methode gibt, um eine erfolgte HIV-Infektion zu heilen, kann die HIV-Pandemie nur gestoppt werden, wenn Neuinfektionen verhindert werden. Obwohl bereits diverse Impfstoffkandidaten für einen HIV-Impfstoff in klinischen Studien getestet wurden, konnte bisher in den Proband*innen keine schützende Immunantwort im ausreichenden Maße stimuliert werden. Das Hauptziel der derzeitigen Anstrengungen ist es, eine potente neutralisierende Antikörperantwort gegen HIV zu induzieren. In spite of intense global research efforts, a vaccine against the human immunodeficiency virus (HIV) is not available to date. In 2019, 38 million individuals were living with an HIV infection, which, if left untreated, causes the deadly acquired immunodeficiency syndrome (AIDS). The development of an HIV vaccine is particularly challenging because HIV can use a multitude of resistance mechanisms to distract the immune system and avoid recognition. For example, a dense coat of host-derived sugars (= glycans) covers the envelope protein of HIV, the sole possible target for the immune system on the viral surface, making the protein appear to be part of the host’s body. While the so-called glycan shield is very effective in sabotaging the recognition of HIV by antibodies, in rare cases infected individuals develop antibodies that break through or directly bind to the glycan shield. In my research group, we use modern technologies to i) develop immunogens to induce glycan-binding antibodies, ii) explore how viral escape from glycan-binding can be avoided, and iii) profile the neutralizing and non-neutralizing functions of glycan-binding antibodies in patient samples to inform new vaccine strategies.
