Synthese von Ahp-Cyclodepsipeptiden als Inhibitoren für Serinproteasen

Eine Fehlregulation von Serinproteasen ist die molekulare Basis bzw. beeinflusst eine Vielzahl von Erkrankungen wie Arthritis, altersbedingte Makuladegeneration, Parkinson oder Alzheimer. Hemmstoffe dieser Proteasen können dementsprechend vielversprechende Wirkstoffe darstellen. Zur Hemmung von Serinproteasen werden bisher Moleküle aus unterschiedlichsten Verbindungsklassen eingesetzt; viele dieser Verbindungen haben jedoch nur begrenzte Hemmpotenz oder -selektivität. Deshalb ist immer noch eine Entwicklung neuer Inhibitoren bzw. Inhibitorklassen nötig, um die Selektivität und das Inhibitionspotential zu steigern, sowie die Zugänglichkeit zu Wirkstoffen für speziellere Serinproteasen zu ermöglichen. Häufig werden Naturstoffe als Grundgerüst verwendet, welche bei einem Screening inhibitorische Eigenschaften zeigten.[8] Die Verbindungen der Naturstoffklasse der Ahp-Cyclodepsipeptide wechselwirken sowohl mit den S- also auch mit den S'-Taschen der Serinproteasen und stellen potente Inhibitoren ausgewählter Serinproteasen wie z. B. Elastase, Chymotrypsin oder Trypsin dar.[9] Die Inhibition anderer Serinproteasen wurde bisher nicht gezeigt. Obwohl bereits Syntheserouten zu diesen Naturstoffen in der Literatur berichtet wurden, sind alle diese Synthesen vergleichsweise aufwendig und unpraktisch und ermöglichen nur begrenzt die Aufstellung medizinalchemisch-relevanter Struktur-Wirkungsbeziehungen.[12,14,119,134] Dementsprechend wurde im ersten Teil der Arbeit die Entwicklung einer neuen praktikablen Synthesestrategie für Ahp-Cyclodepsipeptid durchgeführt. Diese wurde anhand des Naturstoffes Tasipeptin A etabliert. Die etablierte Strategie ist dabei eine Mischung aus Flüssig- und Festphasensynthese. Als Ausgangspunkt wurde eine Glutaminsäure verwendet, deren Aminogruppe durch Fmoc und deren Carbonsäure als Allylester geschützt war. Durch die Bildung eines gemischten Anhydrids und anschließender Behandlung mit Natriumborhydrid konnte die freie Seitenkette zum Alkohol reduziert werden. C- und N-Terminus waren weiterhin geschützt und das 5-Hydroxynorvalin wurde mit Hilfe von Pyridin auf der festen Phase verankert, wobei 2-Chlorotritylchlorid als Linker verwendet wurde. Mit der Fmoc-Strategie wurden die Peptidfragmente in der Reihenfolge Valin, Threonin, Valin und Butansäure gekuppelt. Als Aktivierungsreagenzien wurde HBTU/HOBt und zum Abspalten der Fmoc-Schutzgruppen Piperidin verwendet. Im nächsten Schritt konnte der ungeschützte sekundäre Alkohol des Threonins unter DMAP-Katalyse und Valin-Aktivierung mit DIC verestert werden. Nach dem Kuppeln eines N-methylierten Phenylalanins wurde Fmoc-Leu-OH mit PyBrOP aktiviert und als letzte Aminosäure gekuppelt. Fmoc wurde entfernt und der Allylester mit Hilfe eines Palladium-Katalysators und Morpholin gespalten. Durch eine Festphasen-Macrolactamisierung wurde der Zyklus erhalten, welcher mit Trifluoressigsäure von der festen Phase abgespalten wurden. Der freie Alkohol des Zyklus wurde in Lösung mit Dess-Martin-Periodinan zum Aldehyd oxidiert, welcher das Hemiaminal, die Ahp-Einheit, bildete. Tasipeptin A konnte mit einer Ausbeute von 1.9 % synthetisiert werden. Durch die biologische Evaluierung konnte gezeigt werden, dass nicht nur Elastase, sondern auch zum ersten Mal mit dieser Verbindungsklasse die humanen HTRA2 und -3-Proteasen gehemmt werden können. Im zweiten Teil der Arbeit wurde auf Basis des Ahp-Cyclodepsipeptides eine Leitsequenz entwickelt, mit der es möglich war, auch HTRA1 zu hemmen, dessen Hyperaktivität Krankheiten wie Arthritis oder altersbedingte Makuladegeneration verursacht. Um spezifische Einflüsse von Positionen im Molekül auf die Inhibition zu untersuchen, wurden ein Phenylalanin-Scan durchgeführt und weitere Derivate synthetisiert. Durch das Einführen eines Phenylalanins (20) in die P2' Position konnte eine Verbesserung der Selektivität gegenüber HTRA1 im Vergleich zu Elastase beobachtet werden. Wird das Threonin in Position P2 durch Hydroxy-Phenylalanin (16) oder Hydroxyleucin (21) ersetzt, kann HTRA2 im nanomolaren Bereich inhibiert werden. Wird Phenylalanin in die P3-Position eingeführt (17), sind ähnliche Ki-Werte bestimmt worden. Um inhibitorische Effekte zu verbessern, scheint eine einfache Kombination von Derivaten nicht möglich zu sein. Durch das Einführen eines Hydroxyleucins in Position P2 und eines Phenylalanins in P3 (24) konnte lediglich Elastase inhibiert werden. Die Besetzung der S1-Tasche spielt bei Serinproteasen eine entscheidende Rolle.[15] Um sowohl das Inhibitionspotential also auch die Selektivität zu verbessern, wurden weitere Derivate synthetisiert, bei denen systematisch die P1-Position verändert wurde. Entsprechend der Literatur wurden Aminosäuren mit kleinen hydrophoben Seitenketten ausgewählt, die sowohl zyklische als auch aliphatische Reste aufwiesen.[7,15] Systematisch wurden die Seitenketten verlängert, verzweigt, sowie Doppel- und Dreifachbindungen eingefügt. Die synthetisierte Verbindung 32 trägt ein Cyclopropylglycin in der P1-Position und ist die erste, die eine höhere Affinität gegenüber HTRA1 im Vergleich zu Elastase zeigt. Beide können im niedrigen mikromolaren Bereich gehemmt werden. Um die Inhibition und die Selektivität weiter zu verbessern, wurden auf dieser Grundlage neue Moleküle synthetisiert. Molekulare Docking-Experimente zeigten, dass das Einführen von polaren Resten in die P2'-Position günstig ist. So wurde unter anderem ein Derivat 39 mit einem 4-Chloro-Phenylalanin-Rest synthetisiert. Diese Modifikation verbessert die Inhibition von HTRA1 signifikant und konnte mit einem Ki-Wert von 0.065 µM in den nanomolaren Bereich verschoben werden. Im Rahmen dieser Arbeit konnte eine effektive Fest-Flüssigphasensynthese für Ahp-Cyclodepsipeptide entwickelt werden, welche auch funktionelle Gruppen toleriert. Durch die Synthese von 29 Derivaten und deren biologischer Evaluierung konnten wichtige Informationen über die Inhibitionseigenschaften dieser Naturstoffklasse gewonnen werden. Gleichzeitig konnte gezeigt werden, dass es möglich ist, maßgeschneiderte Inhibitoren zu synthetisieren. Zudem konnten die bisher potentesten, nicht-kovalenten Inhibitoren für HTRA1 und HTRA2 entwickelt werden. In Zukunft sollten weitere Positionen des Moleküls systematisch verändert werden, um die Selektivität und die Inhibition zu verbessern. Durch das Aufnehmen einer Kristallstruktur eines Ahp-Cyclodepsipeptid-HTRA1-Komplexes könnten Wechselwirkungen noch besser nachvollzogen und Derivate zielgerichteter synthetisieren werden. Ein anderer Ansatz ist die Modifikation der Seitenkette des Zyklus. Diese endet mit einem N-Terminus. Würde die Synthese der Seitenkette mit einer Dicarbonsäure beginnen, so wird die Abfolge von C- ' N-Terminus zu N-' C-Terminus geändert werden. Die Seitenkette endet dann mit einer Carbonsäure, welche wiederum an die PDZ-Domäne der HTRA1-Protease binden kann.[7] So ergäbe sich ein „bivalenter" Inhibitor, welcher voraussichtlich selektiver und potenter wäre.

Hyperactivity of serine proteases is involved in several severe human disorders such as arthritis, age-related macular degeneration, Parkinson's or Alzheimer's disease, suggesting that molecular inhibition of serine proteases may represent a promising approach to combat these diseases Accordingly, a wide variety of compound classes for serine protease inhibition have been developed in the last decades; however, despite years of research, many of them still harbor disadvantages such as low inhibition potency or selectivity. Accordingly, the development of new inhibitors and inhibitor classes for improving inhibition selectivity and potency as well as to generate customized inhibitors for specific serine proteases is still a scientifically challenging task. To this end, natural products with suitable inhibitory properties are frequently used as a scaffold for compound development.[8] The the natural product class of Ahp-cyclodepsipeptides act as non-covalent serine proteases inhibitors. They interact with the S- and the S 'pockets, and thus show a high inhibition potential vs. various serine proteases such as elastase, chymotrypsin, and trypsin. Although several synthetic approaches to this compound class have been reported in the last years, all of them are rather complicated and impractical for medicinal chemistry purposes. Accordingly, the first part of this work deals with the development of an alternative practical synthesis strategy for generating Ahp-cyclodepsipeptides. To this end, a chemical synthesis of the natural product Tasipeptin A was established. The resulting strategy turned out to be a mixture of liquid and solid phase synthesis. Glutamic acid was thereby used as a starting point for the synthesis. To this end, the amino group of glutamic acid was protected by Fmoc while the carboxylic acid residue was protected as an allyl ester. The formation of a mixed anhydride and subsequent treatment with sodium borohydride then reduced the free side chain carboxylic acid to an alcohol moiety. As the protecting groups on the C and N terminus were maintained, the 5-hydroxynorvaline residue could subsequently be anchored on the 2-chlorotrityl resin via its OH group using pyridine as a base. By usage of the Fmoc SPPS strategy, the amino acids valine, threonine, valine and butyric acid were subsequently coupled to the resin, using HBTU/HOBt as the activation and piperidine as the Fmoc deprotection reagents. In the next step, the unprotected secondary alcohol of threonine was esterified with valine via DMAP catalysis and DIC activation. After coupling of an N-methylated phenylalanine residue, Fmoc-Leu-OH was coupled after PyBrOP activation. Fmoc was removed and the allyl ester cleaved with morpholine under palladium catalysis. Solid-phase macrolactamization, followed by cleavage from the solid phase with trifluoroacetic acid. Resulted in the cyclized alcohol precursor that was oxidized in solution with Dess-Martin periodinane to the aldehyde. This aldehyde spontaneously formed the hemiaminal Ahp unit. Tasipeptin A was synthesized with a yield of 1.9% (based on the initial resin loading). Subsequent biochemical evaluation of its inhibitory properties then demonstrated that this natural product inhibits the human proteases HTRA2 and -3 in the low micromolar range. In the second part of the work, inhibitors of HTRA1 based on Ahp-cyclodepsipeptide structure were developed. HTRA1 hyperactivity causes diseases such as arthritis or age-related macular degeneration. To this end, systematic variations of a starting Ahp-cyclodepsipeptide inhibitor, e.g. via a phenylalanine scan, were performed. These studies revealed that for example introduction of a phenylalanine residue (20) into the P2' position results in an improvement in HTRA1 inhibition selectivity. Substitition of the threonine residue in position P2 by hydroxy-phenylalanine (16) or hydroxyl leucine (21) results in nanomolar HTRA2 inhibitors. Finally, introduction of phenylalanine in the P3 position (17), similar Ki values have been determined. A proper occupation of the S1 pocket plays a decisive role in the inhibition of serine proteases.[15] To improve the inhibition potential and the selectivity of the Ahp-cyclodepsipeptides for HTRA1, further derivatives were synthesized in which the P1 position was systematically altered. According to the literature, amino acids with small hydrophobic side chains with cyclic or aliphatic residues should be beneficial for HTRA1 inhibition.[7,15] Several derivatives with extended, branched and double and triple bond-carrying side chains were incorporated in this position and one derivative, compound 32 with a cyclopropyl residue, was the first derivative that displayed more potent HTRA1 than elastase inhibition; both are inhibited in the low micromolar range. To further improve inhibition and selectivity, molecular docking experiments were then performed that showed that the introduction of polar residues into the P2'-position may further enhance inhibition properties. Accordingly, a set of P-modified derivatives were synthesized and one derivative (39) bearing a 4-chloro-phenylalanine residue at this position displayed potent HTRA1 inhibition in the nanomolar range (with a Ki value of 0.065 μM). In addition, these studies also demonstrated the effectiveness of the established solid-liquid-phase synthesis of Ahp cyclodepsipeptides as these derivatives also harbored chemically more challenging functional groups. The synthesis of overall 29 derivatives and their biochemical evaluation provided important information about the inhibition properties of the Ahp- natural product class and demonstrated that this compound class allows the synthesis of tailor-made inhibitors. In addition, the most potent noncovalent inhibitors of HTRA1 and HTRA2 were developed. In the future, further positions of the Ahp-cyclodepsipeptide scaffold should be systematically altered for further improving inhibition selectivity and potency. These studies could be guided by structural studies from crystallographic analyses of Ahp-cyclodepsipeptide-HTRA1 complexes,. An alternative approach to " better" inhibitors could be introduce PDZ domain binding sequences at the N-terminus of the Ahp-cyclodepsipeptides, thereby enabling interactions of the Ahp-cyclodepsipeptides not only with the protease domain but also with the PDZ domains of HtrA proteases. Such bivalent inhibition modes may not only improve binding but also other pharmacological parameters such as compound solubility[7].
