Prozessintensivierung auf Basis nachwachsender Rohstoffe

  • Die ganzheitliche stoffliche Verwertung von Biertreber in Bioraffinerieprozessen mit dem Ziel der Itaconsäureproduktion, bietet eine alternative Nutzungsmöglichkeit für diesen industriellen Nebenproduktstrom. Dabei stellt Itaconsäure ein nachhaltiges Substitut für beispielsweise erdölbasierte Acrylsäure dar. Das Ziel der vorliegenden Arbeit liegt in der Entwicklung und Etablierung geeigneter mechanischer, hydrothermaler und enzymatischer Vorbehandlungsmethoden, um zuckerreiche Hydrolysate zu Fermentationszwecken bereitzustellen. Die Optimierung der Einflussgrößen auf die hydrothermale Vorbehandlung erfolgte im Rahmen einer statistischen Versuchsplanung. Dadurch wurden mittels der kombinierten Vorbehandlung, bestehend aus mechanischem Abpressen und anschließender Organosolv Behandlung, im enzymatischen Schritt Ausbeuten von bis zu 66 % (Glucose) und 70 % (Xylose) erzielt. Zur anschließenden fermentativen Herstellung von Itaconsäure wurde ein Produktionsprozess mit dem Organismus Ustilago maydis und den zuckerreichen Biertreberhydrolysaten als Fermentationsmedium etabliert. Dabei konnten Ausbeutesteigerungen von bis zu 52 %, verglichen mit einer Referenz in Standardmedium, erzielt werden. Unter Verwendung von biertreberbasierten Fermentationsmedien wurde bei der Herstellung von bis zu 9,4 g/L Itaconsäure die bereitgestellte Menge an Kohlenhydraten als produktbildungslimitierender Faktor identifiziert. Des Weiteren konnte durch Variation der Prozessführung unter anderem die Option der simultanen Verzuckerung und Fermentation von hydrothermal vorbehandeltem Biertreber mit U. maydis erfolgreich demonstriert werden. Abschließend erfolgte die Evaluation des gesamten entwickelten Bioraffinerieprozesses anhand einer Stoffstromsimulation. Dabei zeigte sich, dass neben der Erhöhung der Ausbeuten noch weitere Optimierungen durchgeführt werden müssen, um ein wirtschaftlich tragfähiges Verfahren zu entwickeln.
  • The holistic utilization of brewer's spent grain in biorefinery processes, with the aim of producing itaconic acid, provides an alternative option of valorisation for this industrial by-product. Itaconic acid can be implemented into existing production routes as a sustainable substitute for petrol-based acrylic acid. The aim of the present work is to develop and establish suitable mechanical, hydrothermal and enzymatic pretreatment methods to provide sugar-rich hydrolysates that can be utilized in fermentation processes. The optimization of the parameters having an influence on the hydrothermal pretreatment was supported by statistical experimental design. This resulted in yields of 66 % (Glucose) and 70 % (Xylose) during the enzymatic saccharification, when deploying a combined pretreatment strategy, consisting of a mechanical treatment with a screw press, followed by organosolv treatment. For the subsequent production of itaconic acid in a fermentation process, a production process using the organism Ustilago maydis and fermentation media based on brewer’s spent grain was established. An increase in yield of up to 52 %, compared to a reference fermentation carried out in standard medium, could be achieved. When using fermentation media based on brewer’s spent grain, the amount of provided carbohydrates was identified as the limiting factor in the production of itaconic acid of up to 9,4 g/L. Furthermore, among others the possibility of a simultaneous saccharification and fermentation process with hydrothermally pretreated brewer's spent grain and U. maydis was successfully demonstrated by varying the process layout and combining the enzymatic saccharification and the fermentation process. Finally, an evaluation and discussion of the developed biorefinery process was carried out based on a simulation of the material flows. This revealed that besides increasing yields, further optimizations must be implemented in order to develop an economically viable process.
