  • Due to the Maillard reaction many compounds are generated, that may possess antioxidative or anticarcinogenic activities as well as toxicity. The aim of this study was the characterization of cellular activities of food extracts from traditional Chinese foods such as Peking duck (PE) and Jinhua ham (JS). In these food extracts the food contaminant acrylamide (AA) has been identified. However, the B-vitamin niacin represents a possibility to reduce AA levels in food by vitamin-binding forming an acrylamide-niacin-adduct (ANA). Niacin as well as AA were generated during roasting of coffee, so the formation of ANA in coffee beans during the heating process could be suggest. The generation of ANA during coffee roasting has been approved by LC/MS-analysis. Therefore, intake of ANA is possible and a supplementation with niacin could prove an effective possibility to reduce the AAcontent in food during preparation. Before considering this possibility, studies are necessary to demonstrate the safety of ANA especially in comparison to AA. Due to the structural similarity to N-methylpyridinium (NMP), a well known Nrf2 activator, ANA was expected to activate the Nrf2-signaling pathway, too. Investigations were performed in HT29 cells with a special emphasis on cytotoxicity, anti- and prooxidative effects, genotoxicity and the impact on Nrf2/ARE signaling pathway. In the used test systems, JS hardly showed cellular effects, with the exception of a slight DNA-damaging potential and a stronger effect on the UGT1A1-gene expression, which did not lead to a protection against a menadione-induced oxidative stress. PE increased the level of intracellular ROS as well as the number of (oxidative) DNA-strand breaks. Subsequently a significant increase of the transcription level of UGT1A1 and a strong inhibition of the cell proliferation were observed. Nor a modulation of the Nrf2/ARE signaling pathway neither an antioxidative effect could be determined after a treatment with ANA. However, the binding of AA to the B-vitamin niacin led to an adduct devoid of cytotoxic and DNA-damaging properties, suggesting a potential way to decrease the in vitro genotoxicity of AA.
