Untersuchung von magnetischen Nanopartikeln hinsichtlich ihrer biologischen Verträglichkeit und der Nutzung als Wirkstoffträger

In this work different nanoparticles were tested in vitro for their biocompatibility and for their potential as drug carriers. The nanoparticles consist of magnetite (Fe3O4), cores that are encapsulated in a silica (SiO2) shell. In orthopaedics, implant-associated infections are a major challenge. Their treatment is difficult due to an insufficient drug concentration in the infection-area. In a system called “Implant-Directed Magnetic Drug Targeting”, nanoparticles are guided by magnetic forces and carry the pharmaceuticals right into the infection-area with the aim to improve the treatment of peri-implant injections. During the experiments the tested nanoparticles showed biocompatibility with different cell lines as well as primary immune cells and osteoblasts. The cell uptake of nanoparticles was shown for different cell types but not for endothelial cells. The nanoparticles could be loaded with 120 µg of enrofloxacin per mg of nanoparticles. After 6 hours about 75 % of the drug was released. First, 3 different nanoparticles were tested for their influence on cell proliferation and viability of a murine fibroblast cell line NIH-3T3 and of a human hepatoma cell line HepG2. The nanoparticles differed in their size (about 110 nm and 60 nm) and in the additional functionalisation with polyethylenglycol and the fluorescent dye rhodamine-B-isothiocyanate. For all 3 nanoparticles no negative influence on proliferation and viability was shown after 24 and 48 h. In the next step murine primary cells were generated: Two cell types of the innate immune system (dendritic cells and macrophages) and bone cells (osteoblasts). The immune cells show no influence on proliferation after 7 days of treatment. The viability was slightly decreased after 3 days. Dendritic cells, macrophages and osteoblasts did not produce proinflammatory cytokines as a reaction to the nanoparticle treatment. Also, the migration of dendritic cells was not influenced. After 21 days of treatment osteoblasts showed no influence on viability or calcium phosphate production. It was shown, that macrophages phagocytise nanoparticles and that afterwards they can be pulled with a magnet. Thanks to staining techniques for the cell nucleus and cytoskeleton you could see that nanoparticles with fluorescent dyes were internalised after 24 h into the cytoplasm of cells without the ability to phagocytise. But there was no internalisation of nanoparticles into humane endothelial cells. Two different nanoparticles were analysed for their drug load capacity. The results show that they could be loaded with 100 – 120 µg enrofloxacin per mg of nanoparticles. After 30 min about 30 %, after 6 h about 75 % of the total drug load of enrofloxacin was released. Taking everything into consideration, the nanoparticles seem to be in vitro biocompatible for different cell types and they can be loaded with high amounts of enrofloxacin. Thus, they can be recommended for additional experiments in vivo.

Im Rahmen dieser Arbeit wurden verschiedene Nanopartikelchargen in vitro auf ihre Biokompatibilität und ihre Eignung als Wirkstoffträger hin untersucht. Im Grundaufbau handelte es sich um Nanopartikel mit einem Kern aus Magnetit (Fe3O4), der mit einer Schicht von Silica (SiO2) ummantelt wurde. Die Nanopartikel sollen als Wirkstoffträger im Bereich des Implant-Directed Magnetic Drug Targeting die Behandlung von peri-implantären Infektionen verbessern, indem sie die Wirkstoffe über magnetische Kräfte gezielt im Infektionsort bringen. Die getesteten Nanopartikel zeigten bei den Versuchen eine Biokompatibilität mit verschiedenen Zelllinien, sowie primären Immunzellen und Osteoblasten. Die Internalisierung von einzelnen Nanopartikeln konnte zwar für verschiedene Zelltypen gezeigt werden, jedoch nicht für Endothelzellen. Die Nanopartikel konnten mit etwa 120 µg Enrofloxacin pro mg Nanopartikelpulver beladen werden und setzten davon nach 6 h etwa 75 % frei. Im ersten Schritt wurden 3 verschiedene Nanopartikel auf ihren Einfluss auf die Proliferation und Vitalität der murinen Fibroblastenzelllinie NIH-3T3 und der humanen Hepatomazelllinie HepG2 hin untersucht. Die Nanopartikel unterschieden sich in der Größe (etwa 110 nm und 60 nm) und in einer zusätzlichen Funktionalisierung mit Polyethylenglycol und dem Fluoreszenzfarbstoff Rhodamin-B-Isothiocyanat. Für alle 3 Nanopartikel konnte keine negative Beeinflussung der Proliferation und Vitalität nach 24 und 48 h gezeigt werden. Im nächsten Schritt wurden aus Mäusen Primärzellen gewonnen. Es handelte sich um 2 Zelltypen des angeborenen Immunsystems (dendritische Zellen und Makrophagen), sowie um Knochenzellen (Osteoblasten). Die Immunzellen zeigten nach 7 Tagen Behandlung mit Nanopartikeln eine unbeeinflusste Proliferation. Die Vitalität wurde nach einer dreitägigen Nanopartikelbehandlung zum Teil schwach herabgesetzt. Dendritische Zellen, Makrophagen und Osteoblasten zeigten keine vermehrte Produktion von proinflammatorischen Zytokinen (TNF-α, IL-6) als Reaktion auf eine Behandlung mit Nanopartikeln. Auch die Migration von dendritischen Zellen war unbeeinflusst. Bei Osteoblasten zeigte sich nach 21 Tagen keine Beeinflussung der Vitalität oder der Calciumphosphatproduktion. Es wurde gezeigt, dass Makrophagen die Nanopartikel phagozytieren und sich die Zellen dann mit einem Magneten anziehen lassen. Durch Anfärbungen des Zellkerns und des Zellzytoskletts konnte beobachtet werden, dass einzelne Nanopartikel, an die Fluoreszenzfarbstoffe gebunden wurden, auch von phagozytoseunfähigen Zellen nach 24 h aufgenommen werden. Jedoch konnte keine Internalisierung von Nanopartikeln in humane Endothelzellen gezeigt werden. Für die Wirkstofffreisetzung von Enrofloxacin aus Nanopartikeln wurden zwei verschiedene Chargen untersucht. Den Untersuchungen nach, konnten sie mit etwa 100 – 120 µg Enrofloxacin pro mg Nanopartikelpulver beladen werden. Nach 30 min wurden etwa 30 %, nach 6 h etwa 75 % der Gesamtmenge an Enrofloxacin freigesetzt. Insgesamt gesehen, scheinen die getesteten Nanopartikel in vitro biokompatibel für verschiedene Zelltypen und können dazu mit hohen Mengen an Enrofloxacin beladen werden. Somit können sie für weitere Experimente in vivo empfohlen werden.
