Aufbau eines Prüfmodells zur in vitro Testung der Anti-Plaque- Wirksamkeit von antimikrobiellen Substanzen

  • Bakterien, die in einem Biofilm organisiert sind, weisen eine um ein Vielfaches höhere Resistenz gegenüber antimikrobiellen Substanzen auf als planktonisch gelöste Zellen. Der häufigste beim Menschen vorkommende Biofilm ist die dentale Plaque. Als Keim zur Herstellung eines Biofilms wurde Streptococcus sanguis benutzt. Dieser bildete nach 48 h aerober Bebrütung bei 37 °C einen sichtbaren Biofilm auf Hydroxylapatit-Plättchen, die zur Imitation der Zahnoberfläche in einer Wachstumskammer mit kontinuierlicher Flusskultur-Technik aufgehängt waren. Zur Überprüfung der Funktionalität des Modells wurde die Wirksamkeit von drei Antiseptika getestet. Die HA-Plättchen wurden aseptisch aus der Wachstumskammer entnommen und jeweils in verschiedene Reagenzröhrchen mit Chlorhexidin (0,1% oder 1,0%), PVP-Iod (1,5% oder 7,5%) sowie Octenidindihydrochlorid (0,05% oder 0,1%) gegeben. Die Einwirkzeit jeder Konzentration betrug 5 min oder 30 min. Proben aus der Bakteriensuspension der Wachstumskammer wurden entsprechend behandelt. Ein zugefügtes spezifisches Neutralisationsmittel beendete die Wirkung der Antiseptika. Es lag eine signifikante Differenz zwischen der antimikrobiellen Aktivität gegen Bakterien in gelöster Form und solchen in Biofilmen vor. Beste Reduktionsfaktoren konnten mit Chlorhexidin (1,0%, 30 min), sowohl in Bezug auf Biofilme (3,97 log) als auch auf planktonische Zellen (= 5,58 log), ermittelt werden. Bei jeder der getesteten Substanzen zeigte sich eine klare Dosis-Zeit-Wirkungs-Beziehung. Es wurde daher geschlussfolgert, dass das entwickelte Modell in der Lage ist, schnell und kosteneffektiv die Aktivität antimikrobieller Substanzen gegen als Biofilm gewachsene Bakterien darzustellen.
  • Bacteria grown in biofilms are less susceptible to antimicrobial agents than planktonic bacteria. One of the most common biofilms in humans is dental plaque. To investigate the antimicrobial activity against dental bacteria grown in biofilms, we used Streptococcus sanguis, which forms a biofilm on hydroxyapatite disks, used to mimic the tooth surface. After 48 h of aerobic incubation at 37 °C in a continuous flow culture, a biofilm consisting of S. sanguis became visible on the surface of the disks. Screening the functionality of the model three antimicrobial agents were tested. The disks were aseptically removed from the growth chamber and placed in different vessels containing either chlorhexidine (0.1% or 1.0%), PVP-iodine (1.5% or 7.5%), or octenidindihydrochloride (0.05% or 0.1%) for 5 or 30 min. In addition, the antiseptics were applied to the bacterial suspension in the growth chamber. To stop the effect of the antimicrobial agents a specific neutralizing solution was added. A significant difference was observed in the antimicrobial activity against bacteria in the suspension liquid compared to the bacteria grown in biofilms. The best reduction factors were obtained with chlorhexidine (1.0 %, 30 min) for either sessile (3.97 log) or planktonic bacteria (= 5.58 log). Clear relationships between the doses and times of action were found for the assessed agents. Therefore, it can be concluded that the present in-vitro assay is a quick and cost-effective model to screen the activity of antimicrobial agents against bacteria grown in biofilms.
