Synthese 1,5-substituierter Benzimidazolone und Benzimidazolthione sowie N-substituierter 7-Chlorchinolin-4-amine zur Untersuchung ihrer biologischen Aktivität

  • Die Bekämpfung der „most neglected diseases“ stellt die Menschheit vor eine große Herausforderung. Besonders betroffen sind Menschen in den Ländern der Dritten Welt. Zur Behandlung vieler dieser Erkrankungen gibt es bis jetzt noch keine ausreichend aktiven Arzneistoffe. Außerdem stellt die Resistenzentwicklung der Erreger gegen vorhandene Antiinfektiva ein äußerst großes Problem dar. Gegenstand dieser Arbeit war die Synthese von Substanzen, die als Inhibitoren der tRNA-Guanin-Transglycosylase (TGT) untersucht werden sollten und solche, die auf ihre antiplasmodiale Aktivität in vivo getestet werden sollten. Das Target des TGT-Projekts, die tRNA-Guanin-Transglycosylase von Shigella-Spezies, ist ein bakterielles Enzym, das verantwortlich für Pathogenitätsmechanismus virulenter Shigellen ist. Die TGT hat eine Schlüsselfunktion bei der Expression von Virulenzfaktoren der Shigellen und spielt somit eine wichtige Rolle bei der Entstehung der Bakterienruhr (Shigellose). Sie katalysiert den Austausch der Purinbase Guanin durch die modifizierte Base preQ1 in der Anticodon-Wobble-Position bakterieller tRNA. Bei Untersuchungen von Shigellen mit mutiertem TGT-Gen konnte eine signifikante Abnahme der Pathogenität dieser Erreger festgestellt werden. Dieses Enzym kann deshalb prinzipiell als mögliches Target für neuartige biologisch aktive Substanzen betrachtet werden, denn die Hemmung der TGT führt zum Verlust der Pathogenität der Erreger, allerdings nicht zu deren Tod. Im Rahmen der vorliegenden Arbeit wurden Abkömmlinge der Leitstruktur 5-Nitro-1,3-dihydro-2H-imidazo[4,5-b]pyridin-2-on, die aus einem virtuellen In-silico-Screeningexperiment hervorgegangen ist, synthetisiert um diese in einem Radioligandenassay auf TGT-Inhibition zu untersuchen. In vorangegangenen Untersuchungen konnte gezeigt werden, dass der Austausch des Pyridinstickstoffatoms der Leitstruktur durch ein Kohlenstoffatom zu einer Verbindung mit vergleichbarer inhibitorischen Aktivität führt. Ausgehend von dieser veränderten Leitstruktur wurden Benzimidazol-2-one und -thione synthetisiert. Diese Verbindungen weisen unterschiedliche Gruppen in Position 5 auf und unterscheiden sich zudem durch die Substitution an Position 1. Die Synthese der Verbindungen mit Benzimidazol-Grundkörper erfolgte durch die initiale Einführung der Substituenten durch nukleophile Substitution an ortho-Nitro-Halogenbenzen-Derivaten. Für diesen Reaktionsschritt erwies sich die Synthese im Mikrowellenreaktor als besonders günstige Methode. Die Reduktion der entstandenen 2,4-Dinitroanilin-Derivate erfolgte selektiv unter Verwendung von Natriumsulfid als Reduktionsmittel, wobei die ortho-Nitrogruppe unverändert blieb. Die Anilinderivate mit Cyano- und Trifluormethylgruppe in para-Position wurden durch eine palladiumkatalysierte Hydrierung zu ortho-Phenylendiaminen reduziert, die anschließend ohne Isolierung dem Ringschluss unterzogen wurden. Die Ringschlussreaktionen erfolgten mit Triphosgen oder Kohlenstoffdisulfid zu Benzimidazol-2-onen beziehungsweise -thionen. Eine Auswahl an hergestellten Derivaten konnte hinsichtlich ihrer Aktivität als TGT-Inhibitoren untersucht werden. Einige Verbindungen wurden auf ihre Zytotoxizität an vier Krebszelllinien hin untersucht. Das zweite Teilgebiet der vorliegenden Arbeit beschäftigt sich mit N-substituierten 7-Chlorchinolin-4-aminen, die auf ihre antiplasmodiale Aktivität hin untersucht werden konnten. Diese Verbindungen wurden in einer Zweistufensynthese mit abschließender säulenchromatographischer Reinigung synthetisiert. Verbindungen, die in vorangegangenen In-vitro-Untersuchungen eine Aktivität gegen chloroquinresistente Plasmodium-falciparum-Stämme zeigten, konnten in vivo an Mäusen getestet werden. Dabei zeigten einige Vertreter guten Aktivitäten gegen den eingesetzten Erreger.
  • The fight against the “most neglected diseases” is a great challenge for mankind, mainly affecting those living in 3rd World countries. For a lot of these diseases, no adequate effective drugs have been invented so far. Also, the emergence of resistant pathogens against existing antiinfectives constitutes a great problem. The subject of this work has been the synthesis of substances that were examined as inhibitors of tRNA guanine transglycosylase (TGT) in a radioligand binding assay and those that have been tested on their antiplasmodial activity in vivo. The target of the TGT-Project, tRNA guanine tansglycosylase of Shigella species, is a bacterial enzyme that is the cause of the pathogenicity of virulent shigella. In that respect, TGT is crucial in the expression of virulence factors of shigella and as such plays an important role in the development of shigellosis. TGT catalyzes the base exchange of guanine by the modified base preQ1 in the anticodon wobble position of bacterial tRNA. The analysis of shigella with mutated TGT-genes showed significant reduction of the pathogenicity of microorganisms. This enzyme can be considered a possible target for bioactive substances, because TGT inhibition leads to a loss of pathogenicity of shigella but not to their death. Within the framework of this thesis, descendants of the lead 5-nitro-1,3-dihydro-2H-imidazo[4,5-b]pyridin-2-one resulting from an in silico virtual screening experiment, were synthesized to determine their activity as TGT inhibitors in a radioligand assay. Previous studies have shown that the exchange of pyridine nitrogen atom within the lead structure by a carbon atom leads to a compound with comparable inhibitory activity. Based on this altered lead, benzimidazol-2-ones and benzimidazole-2-thiones were synthesized. These compounds exhibit different groups in position 5 and differed through substitution on position 1 as well. The synthesis of compounds with benzimidazole scaffold was done through the initial introduction of substituents by nucleophilic substitution at ortho-nitro halogenbenzene derivatives. For this reaction step, the synthesis in a microwave reactor proved to be a good method. The reduction of the 2,4-dinitroaniline derivatives could be achieved selectively using sodium sulfide as reducing agent, retaining the para-nitro group. The aniline derivatives with cyano- and trifluoromethyl groups in para-position were reduced to ortho-phenylenediamines by palladium-catalyzed hydrogenation. Following, these were subject to cyclization without isolation. The Cyclization reaction was carried out with triphosgene or carbon disulfide towards benzimidazol-2-ones or benzimidazole-2-thiones, respectly. A selection of compounds was analyzed in regard to their activity as TGT inhibitors. Some compounds were analyzed on their cytotoxicity using four cancer cell lines. The second part of the work deals N-substituted 7-chloroquinoline-4-amines that were analyzed for their antiplasmodial activity. These derivatives were synthesized in a two stage synthesis with terminal cleaning by column chromatography. Compounds that showed an activity in previous in vitro analyses against chloroquine resistant Plasmodium falciparum stems, were tested in vivo in mice. Here, some of the compounds showed activity against the pathogenic agent.
