Untersuchungen zur Hemmung der Adenovirusreplikation durch modifizierte Nukleosidanaloga auf zellulärer und molekularer Ebene.

  • Adenovirus-assoziierte Infektionen führen in immunkompetenten und immunsupprimierten Patienten zu signifikanter Morbidität und Mortalität. Bisher gibt es keine effektive antivirale Chemotherapie. Die inhibitorische Aktivität von 20 strukturmodifizierten Nukleosidanaloga gegenüber ADV 7 und ADV 19 auf zellulärer Ebene wurde mittels Fluoreszenzfokusreduktionsassay untersucht. Starke selektive Hemmer der ADV-Replikation waren die Substanzen 2',3'-Didesoxyadenosin (ddA), 2',3'-Didesoxycyitidin (ddC), 3'-Azido-2'-desoxyadenosin (N3AdR), 3'-Fluor-2'-desoxythymidin (FTdR) und 3'-Fluor-2'-desoxyguanosin. Gegenüber ADV 7 waren die Substanzen FTdR (IC50 6,1 µM), NsAdR (IC50 4,75 µM), ddC (IC50 3,1 µM) ddA (IC50 2,8 µM) und gegenüber ADV 19 die Substanzen FTdR (IC50 1,3 µM), FGdR (IC50 1,0 µM) und ddC (IC50 5,5 µM) die effektivsten antiviralen Substanzen. Die antivirale Wirkung der Nukleosidtriphosphatanaloga auf die ADV-Polymeraseaktivität auf molekularer Ebene wurde mittels ADV-Polymeraseassay untersucht. Die Synthese der ADV-Polymerase erfolgte unter Nutzung eines rekombinanten Ad pol Baculovirus Systems. Spodoptera frugiperda Zellen wurden mit rekombinantem Acpol 14 A infiziert um ausreichende Mengen ADV-Polymerase zu weiteren in-vitro-Untersuchungen zu erhalten. Sechs Nukleosidtriphosphat-Analoga wurden evaluiert. Die ermittelten Konzentrationen zur Hemmung der ADV DNA-Polymeraseaktivität um 50 % (IC50) lagen im Bereich von 0,63 bis 2,96 µM. Wirksamste Substanzen waren FTdR, FGdR und ddC.

There is currently no licensed chemotherapy effective in dealing with this virus infection. The anti-adenoviral activity of 20 modified nucleoside analogs against ADV 7 and ADV 19 was investigated on cellular level using Fluorescent focus reduction assay. The compounds 2',3'-Dideoxyadenosin (ddA), 2',3'-Dideoxycyitidin (ddC), 3'-Azido-2'-deoxyadenosin (N3AdR), 3'-Fluor-2'-deoxythymidin (FTdR) and 3'-Fluor-2'-deoxyguanosin emerged as potent and selective inhibitors of ADV replication. Against ADV 7 the compounds FTdR (IC50 6,1 µM), N3AdR (IC50 4,75 µM), ddC (IC50 3,1 µM) ddA (IC50 2,8 µM) and against ADV 19 the compounds FTdR (IC50 1,3 µM), FGdR (IC50 1,0 µM) and ddC (IC50 5,5 µM) where the most effective inhibitors. To investigate the effect of modified nucleoside triphosphate analogs on ADV polymerase activity on molecular level, an assay based on recombinant baculovirus was used. Spodoptera frugiperda cells were infected with the recombinant Acpol 14 A to produce sufficient quantities of ADV polymerase for in vitro studies. Six nucleoside triphosphate analogs where evaluated. The concentrations of nucleoside triphosphate analogs required to inhibit the ADV DNA polymerase by 50% (IC50) ranged from 0,63 to 2,96 µM. The most effective compounds where FTdR, FGdR and ddC.
