RNA-Edierung in Mitochondrien höherer Pflanzen

Zusätzlich zu den bereits bestehenden in organello Systemen für Z. mays und S. bicolor wurden in dieser Arbeit in organello Systeme für die dikotylen Pflanzen B. oleracea var. botrytis und A. thaliana etabliert. Mittels der in organello Systeme wurde die Edierung und das Spleißen von fremden Transkripten in isolierten Mitochondrien analysiert. Es wurde gezeigt, dass Mitochondrien aus B. oleracea var. botrytis und A. thaliana bzw. aus Z. mays in der Lage sind das cox2 Transkript aus Z. mays bzw. aus A. thaliana korrekt zu spleißen und effizient zu edieren. Unter den edierten Positionen der cox2 Transkripte befinden sich auch Edierungspositionen, die im jeweiligen endogenen cox2 Transkript nicht vorkommen. In einer Datenbankanalyse der cox2 RNA-Edierungspositionen mono- und dikotyler Pflanzen wurden zwei Positionen ausschließlich in monokotylen Pflanzen und eine Position nur in dikotylen Pflanzen gefunden. Die Edierung auch dieser Positionen in den heterologen isolierten Mitochondrien führte zu dem sogenannten „self-guiding-transcript” Modell der RNA-Edierung in pflanzlichen Mitochondrien. Dieses Modell besagt, dass die Edierungspositionen im Transkript mittels der Sekundär- oder Tertiärstruktur der RNA von Edierungsfaktoren erkannt und ediert werden. Z. mays cox2 Edierungspositionen wurden durch Punktmutationen in das cox2 Transkript aus A. thaliana eingeführt. Diese Positionen wurden in Mitochondrien aus Z. mays ediert, ebenso wie eine erwartungsgemäße Edierung der A. thaliana cox2 Edierungspositionen stattfand. Mittels Elektroporation wurden außerdem plastidäre Z. mays ndhB- und ycf3- Sequenzen in isolierte Mitochondrien aus B. oleracea var. botrytis und Z. mays eingebracht. Eine Analyse der Transkripte ergab, dass diese in den Mitochondrien weder gespleißt noch ediert werden. Auch eine Entfernung des Introns aus dem ndhB Konstrukt führte nicht zur Edierung der Transkripte in den Mitochondrien. Somit scheinen die mitochondrial lokalisierten Spleiß- und Edierungsfaktoren nicht in der Lage zu sein die plastidären Introns und die plastidären Edierungspositionen zu erkennen. Es erfolgte zudem die Entwicklung eines Protokolls zur Isolierung von Edierungsfaktoren mittels der Interaktion Streptavidin-beschichteter Magnetpartikel mit Biotin-markierter RNA bzw. RNA-Protein-Komplexen. Hiermit wurden durch spezifische Bindung an ein Fragment mit Edierungsposition aus dem A. thaliana cox2 Transkript, Proteine aus mitochondrialen B. oleracea var. botrytis und A. thaliana Extrakten isoliert. Die MALDI-TOF/TOF-massenspektrometrische Analyse eines der isolierten A. thaliana Proteine führte zu einem PPR-Protein.

In organello systems for Z. mays and S. bicolor have been established prior to the begin of this study. In this study in organello systems for the dicotyledonous plants B. oleracea var. botrytis and A. thaliana were developed. The in organello systems were used to analyze splicing and editing of transcripts in heterologous mitochondria. It was shown that Z. mays cox2 transcripts were correctly spliced and edited in the isolated mitochondria of B. oleracea var. botrytis and A. thaliana. Similarly, the A. thaliana cox2 transcripts were efficiently spliced and edited in isolated maize mitochondria. Among the analyzed editing sites some sites are not present in the endogenous transcripts of the electroporated mitochondria. In a database search two cox2 RNA editing sites were found to be located exclusively in monocotyledonous plants, whereas one cox2 RNA editing site was found only in dicotyledonous plants. Even these editing sites are edited in the heterologous mitochondria. Those results support a self-guiding-transcript model for RNA editing in plant mitochondria. In this model the editing information is contained in the transcript itself, as the transcript forms a complex secondary or tertiary structure, guiding the editing factors to the editing sites. Z. mays cox2 editing sites introduced into the A. thaliana cox2 transcript by site-directed-mutagenesis were edited in isolated mitochondria from Z. mays. Furthermore the A. thaliana cox2 editing sites were edited as expected. Plastid Z. mays ndhB and ycf3 sequences were introduced into isolated mitochondria of B. oleracea var. botrytis and Z. mays via electroporation. The transcripts of the plastid sequences were neither spliced nor edited in mitochondria. Removal of the intron of the ndhB construct did not induce editing of the transcript in the mitochondria. These results indicate that mitochondrial localized splicing and editing factors are not capable of recognizing plastid introns and plastid editing sites. In addition a protocol for isolation of editing factors was developed, utilizing the interaction of streptavidin-coated magnetic particles and biotin-labeled RNA or RNA-protein complexes. Using this approach, proteins from mitochondrial B. oleracea var. botrytis and A. thaliana extracts were isolated via specific binding to an A. thaliana cox2 RNA fragment containing an editing site. MALDI-TOF/TOF mass spectrometry of an A. thaliana protein resulted in a PPR protein.
