Untersuchung der Umsetzung asymmetrisch methylierter Argininedurch rekombinant hergestellte und native Dimethylarginin-Dimethylaminohydrolase-2

In den letzten Jahren wurden verschiedene Wege zur Therapie pathophysiologisch erhöhter Stickstoffmonoxid (NO)-Konzentrationen entwickelt. NO wird aus L-Arginin über die NO-Synthasen (NOS) hergestellt, deren direkte Hemmung derzeit therapeu-tisch nicht genutzt werden kann. Als Alternative wurde daher die Hemmung der Di-methylarginin-Dimethylaminohydrolase (DDAH) und die Akkumulation ihrer Substrate N ω-Monomethyl-L-arginin (L NMMA) und N ω,N ω'-Dimethyl-L-arginin (ADMA), die endogen die NOS-Enzyme hemmen, interessant. Da nur die mit der DDAH-1 co-lokalisierte neuronale und induzierbare NOS gehemmt werden soll, werden selektive DDAH-1-Inhibitoren benötigt. Für die Analyse der Isoformen-Selektivität bekannter DDAH-Hemmstoffe wurden die humanen DDAH-Isoformen rekombinant in E. coli hergestellt und gereinigt. Um die Aktivität der Proteine zu prüfen, wurden sie mit den postulierten DDAH-Substraten inkubiert. Im Gegensatz zur rekombinanten hDDAH-1 setzte die hDDAH-2 weder L NMMA und ADMA, noch die alternativ getesteten Substrate S-Methyl-L-thiocitrullin (SMTC) und N ω-Amino-L-arginin zu L Citrullin um. Da es nicht immer gelingt humane Proteine in E. coli funktionsfähig herzustellen, konnte nach den Ergebnissen nicht sicher von einem Fehlen endogener Enzymaktivi-tät ausgegangen werden. Allerdings wurde die hydrolytische Aktivität der DDAH-2 bereits zu Beginn dieser Arbeit kontrovers diskutiert. Daher wurden Untersuchungen mit den nativen DDAH-Isoformen in porcinem Gewebe durchgeführt. Mittels Western Blot-Analysen wurden eine unterschiedliche Organverteilung für die DDAH-Enzyme ermittelt und repräsentative Gewebe für die Untersuchung der Aktivität nativer DDAHs identifiziert: Die Schilddrüse für die DDAH-2 und die Niere für die DDAH-1. Bei Inkubation aller Gewebe mit ADMA und der Schilddrüse und Niere mit allen vier Substraten, konnte native DDAH-2, im Gegensatz zur DDAH-1, kein Substrat zu L Citrullin abbauen. Die Ergebnisse der Studien mit nativen und rekombinanten DDAH-Isoformen beleg-ten somit übereinstimmend, dass DDAH-2 keine endogene hydrolytische Aktivität für L-NMMA und ADMA aufweist. Der postulierte Metabolismus ist daher nicht als Aktivi-tätsassay für die DDAH-2 und zur Testung der Isoformen-Selektivität von DDAH-Inhibitoren geeignet. Die physiologische Funktion der DDAH-2 und der Mechanismus über den sie den NO-Stoffwechsel beeinflusst müssen nun weiter erforscht werden.

In the last years, different strategies were developed for the therapy of diseases based on pathophysiologically raised nitric oxide (NO) concentrations. NO is synthe-sized from L-arginine by nitric oxide synthase isoenzymes (NOS). Until today, a direct inhibition of NOS to attenuate pathophysiological NO-concentrations is limited by side effects of NOS-inhibitors. Therefore, the inhibition of dimethylarginin-dimethyl-aminohydrolase (DDAH) followed by an accumulation of its substrates N ω-mono-methyl-L-arginine (L NMMA) und N ω,N ω'-dimethyl-L-arginine (ADMA) which are endogenous inhibitors of all NOS-enzymes was a favored alternative. For therapy, exclusive inhibition of inducible NOS (iNOS) and neuronal NOS (nNOS) is crucial. Hence, due to the colocalisation of nNOS/iNOS with DDAH-1, solely selective DDAH 1-inhibitors are of therapeutic interest. To study the isoform selectivity of known DDAH-inhibitors, human DDAH isoforms are expressed and purified from E. coli. The activity of the enzymes was measured by degradation of the postulated DDAH substrates L-NMMA, ADMA, and alternative substrates S-methyl-L-thiocitrulline (SMTC), and N ω-amino-L-arginine to L citrulline. In contrast to hDDAH 1, neither methylarginines nor alternative substrates were me-tabolized by recombinant hDDAH-2. Expression of nonfunctional human proteins in E. coli is a common problem, thus the observed lack of intrinsic activity could not reliably be interpreted as an actual state of the enzyme. However, recent works have raised doubts concerning the hydrolytic activity of DDAH-2. Therefore, studies on the activity of native DDAH were carried out in eight porcine tissues. Western blot analyses revealed an organ specific expres-sion pattern for both DDAH isoforms in tissues. In thyroid solely DDAH-2 was detected, in contrast in kidney high amounts of DDAH-1 were found. Seven tissues were incubated with ADMA, furthermore thyroid and kidney cytosols were incubated with all DDAH substrates. Again, neither ADMA nor other DDAH substrates were converted to L citrulline by native DDAH-2 while all substrates were metabolized by tissue DDAH-1. Taken together, the results with recombinant and native DDAH isoforms are in agreement and clearly indicate that DDAH-2 has no intrinsic hydrolytic activity for methylarginines. Therefore, the postulated metabolism is unsuitable for activity anal-ysis of DDAH-2 and for investigations on isoform selectivity of DDAH inhibitors. The true physiological function of DDAH-2 and the mechanism of the impact on NO me-tabolism have now further to be elucidated.
