Abstract: Immunologische Prozesse im Rahmen der Heilungsphase nach einem Myokardinfarkt (MI) sind Bestandteil der gegenwärtigen Forschung. Das Ziel dieser Arbeit war es, die Auswirkungen einer verstärkten herzspezifischen Autoimmunantwort auf die Heilungsphase des Myokards nach einem experimentellen MI bei Mäusen darzustellen. Hierzu wurden männliche Balb/c Wildtyp Mäuse entweder mit dem kardialen Myosin oder dem kompletten Herzextrakt eine Woche vor einem experimentellen MI immunisiert. Um zu untersuchen, ob es bei den immunisierten Tieren zu Veränderungen in der Heilungsphase des Myokards kam, wurden an Tag 7 nach dem MI u.a. das Herz und die mediastinalen Lymphknoten untersucht. Hierbei zeigte sich, dass die Tiere der Herzextrakt-Gruppe eine signifikant erhöhte Infarktgröße im Vergleich zur Kontrollgruppe aufwiesen. Außerdem exprimierten die mit Herzextrakt immunisierten Mäuse signifikant erhöhte Level der Gene MMP2, MMP9 und Col1a1 im Vergleich zur Kontrollgruppe, welche das ungünstige kardiale Remodeling induzieren. Die Immunisierung mit Myosin hatte hingegen keinen nachteiligen Effekt auf die Heilungsphase des Myokards.
Immunological processes during the healing phase after myocardial infarction (MI) are components of current research. The aim of this study was to determine which effects an increased heart-specific autoimmune response would have on the healing phase after experimental MI in mice. Therefore we immunised male Balb/c Wildtype mice with cardiac myosin or the complete heart extract one week before experimental MI. In order to analyse whether there might have been differences during the healing phase between the two groups of immunised mice, we investigated the heart and mediastinal lymph nodes on day 7 post-MI. It was shown that the animals that received the heart extract had a significant enlarged infarct size compared to the control group. Furthermore, the data revealed a significantly higher expression level of the genes MMP2, MMP9 und Col1a1 in mice immunised with heart extract compared to the control group. These genes are known to induce adverse cardiac remodeling. Moreover, the immunisation with myosin showed no harmful effect on the healing phase after MI.
