Abstract: Die Arbeit untersucht die Bedeutung multipler Typ-I Signalpeptidasen (Sip- SPasen) bei der im Vergleich zu B. subtilis 168 besonders Proteinsekretions- aktiven Spezies B. amyloliquefaciens. Die Arbeit beschreibt in Ergänzung zu den von Hoang, V. & Hofemeister, J. [(1995) BBA 1269, 64-68] isolierten sipS und sipT Genen zwei weitere sip- Gene sipV(Ba) und sipW(Ba). Ein zu sipU- ähnliches Gen wurde nicht gefunden, was zu Spekulationen über das Fehlen einer SipU- homologen SPase bei B. amyploliquefaciens und mögliche Unterschiede zu B. subtilis hinsichtlich der sip-Genausstattung Anlass gibt. In einer PCR-Studie, wird das Fehlen von SipW, einer zu eukaryotischen (ER-Membran-) SPasen verwandten Sip- Isoform, bei 2 von 12 Bacillus- Stämmen, und zwar in der (Extremophilen-) Gruppe V (bei B. stearothermophilus und Thermoactinomyces vurlgaris), festgestellt. Die Klonierungen bilden die Grundlage für die Konstruktion von Disruptionsmutanten für alle vier Gene und die Expression der verschiedenen SPasen in einer E. coli ts- Mutante und anschließende Komplementationsuntersuchungen sowie die Konstruktion erster hybrider SPasen durch Fusion N- und C- terminaler Sequenzen. Die Komplementationsstudien belegen funktionelle Unterschiede, z.B. kann SipV(Ba) im Unterschied zu SipS(Ba) offensichtlich wegen starker Proteolyse die lepB -Funktion bei E. coli nicht komplementieren. Ausfallfunktionen bei sip- Disruptionsmutanten lassen Typ-spezifische Funktionen erkennen, u.a. für SipT(ba) in der Prozessierung von Sporulationsprotein(en), für SipV(Ba) in Beziehung zum Transport eines unbekannten Autolysins oder von SipS(Ba) sowie SipT(Ba) für den Export einer (unbekannten) 35 kDa- Nuklease. Mit dem in dieser Arbeit als potentiell spezifisches Sip-Target isolierten Chitin-Bindeprotein (ChbB) wird ein für B. amyloliquefaciens bisher unbekanntes Exportprotein beschrieben, welches bei B. subtilis fehlt aber entgegen der Annahme, nicht Sip-spezifisch exportiert wird.
The use of B. amyloliquefaciens for enzyme production and its exceptional high protein export capacity initiated this study where the presence and function of multiple type I signal peptidase isoforms was investigated. In addition to type I signal peptidases SipS(ba) [Meijer et al. (1995) Mol. Microbiology 17, 621-631] and SipT(ba) [Hoang, V. & Hofemeister, J. (1995) BBA 1269, 64-68] which were previously identified, these studies present evidence for two other Sip-like genes in B. amyloliquefaciens. Same map positions as well as sequence motifs verified these genes to encode homologues of SipV and SipW of B. subtilis. SipW-encoding DNA was also evidenced for other Bacillus strains representing different phylogenetic groups. Sequence alignments of 23 known Sip-like proteins from Bacillus origin indicated further branching of the P-group signal peptidases into clusters represented by B. subtilis SipV, SipS-SipT-SipU and B. anthracis Sip3-Sip5 proteins, respectively. Each B. amyloliquefaciens sip(ba) gene was expressed in an E. coli LepBts mutant and tested for genetic complementation of the temperature sensitive (TS) phenotype as well as pre-OmpA processing. Although SipS(ba) as well as SipT(ba) efficiently restored processing of pre-OmpA in E. coli, only SipS(ba) supported growth at TS conditions indicating functional diversity. Changed properties of sip(ba) gene disruption mutants in cell autolysis, motility, sporulation, and nuclease activities seemingly correlate with specificities and /or localisation of B. amyloliquefaciens SipS, SipT and SipV isoforms. This studies also reveal for the first time that Bacillus amyloliquefaciens ALKO 2718 secretes a small protein (ChbB) which is compared to CHB1 and CHB2 of Streptomyces species. The ChbB protein was purified and characterised The chbB and neighbouring genes were isolated and studied. ChbB homologues are shown to be abundant among different B. amyloliquefaciens strains.
