Discovery of the new antimicrobial compound 7-O-malonyl macrolactin A

In this study the production, isolation, chemical and biological characterization of the new compound 7-O-malonyl macrolactin A is reported. The compound is a new member of the macrolactin family of compounds and was produced, in conjunction with macrolactin A and 7-O-succinyl macrolactin A, by a Bacillus subtilis soil isolate. The in vitro activities of the three macrolactins against reference microorganisms and selected clinical multi-resistant gram-positive and gram-negative pathogens were tested by agar diffusion test and liquid microdilution assays and compared to commercial antibiotics. The 7-O-malonyl macrolactin A was the most active compound against staphylococci and enterococci when tested in the agar susceptibility test. Although the new compound showed a bacteriostatic activity when tested in liquid media, a strong inhibition of the bacterial growth was observed particularly with methicillin-resistant Staphylococcus aureus (MRSA) and vancomycin-resistant enterococci (VRE). Kinetics of growth and post-antibiotic effect (PAE) at sub-minimal inhibitory concentrations (Sub-Mic) corroborated the bacteriostatic activity of the 7-O-malonyl macrolactin A. Transmission electron microscopical observations of MRSA and VRE clinical strains, showed an alteration of the replication process in treated bacteria. Further studies should be performed to characterize the mechanism of action of the new compound against multi-resistant gram-positive bacteria and to evaluate its possible efficacy against infections caused by these bacteria.

In dieser Studie wird über die Produktion, Isolierung, chemische und biologische Kennzeichnung des neuen Mittels 7-O-malonyl Macrolactin A berichet. Das Mittel ist ein neues Mitglied der macrolactin Familie, das durch ein Bacillus-subtilis-Bodenisolat zusätzlich zu Macrolactin A und 7-O-succinyl Macrolactin A produziert wurde. Die In-vitro-Aktivität der drei Macrolactine wurden durch den Agar-Diffusionstest und Microerdünnungtest im Vergleich zu kommerziellen Antibiotika gegen Bezugsmikroorganismen und ausgewählten klinisch multi-beständige, grampositive und gramnegative Krankheitserreger geprüft. Das 7-O-malonyl Macrolactin A war im Nährbodenanfälligkeitstest das aktivste Mittel gegen Staphylokokken und Enterokokken. Obgleich das neue Mittel in flüssigen Medien eine bakteriostatische Aktivität aufwies, wurde eine starke Hemmung des bakteriellen Wachstums, besonders mit Methicillin-beständigem Staphylococcus aureus (MRSA) und vancomycin-beständigen Enterokokken (VRE), beobachtet. Kinetikstudien des Wachstums und des Post-antibiotica Effektes (PAE) bei sub-minimal inhibitory concentrations (Sub-Mic) bekräftigte die bakteriostatische Aktivität des 7-O-malonyl Macrolactin A. Elektronmikroskopische Beobachtungen des MRSA und VRE der klinischen Belastungen zeigten eine Änderung des Reproduktionprozesses in behandelten Bakterien. Weitere Studien sollten durchgeführt werden, um die Einheit der Aktivität des neuen Mittels gegen multi-beständige, grampositive Bakterien zu kennzeichnen und seine mögliche Wirksamkeit gegen die Infektion auszuwerten, die durch diese Bakterien verursacht werden.
