Screening-Strategien für mikrobielle Hochleistungsstämme am Beispiel der 1,3-Propandiol-Produktion aus Rohglycerin

Ziel der Arbeit war die Entwicklung einer maßgeschneiderten Screening-Strategie für die Isolierung von 1,3-Propanidol-Hochleistungsproduzenten. Hierfür sollte ein automatisierbares Hochdurchsatz-Verfahren entwickelt werden. Basierend auf einer konventionellen Batch-Anreicherungsphase für anaerobe, Glycerin-verwertende Mikroorganismen aus Umweltproben wurden zunächst Durchsatz und Selektivität erhöht. Durch Einsatz hoher Konzentrationen an 1,3-Propandiol (1,3-PD) und Rohglycerin sowie verschiedener Nährstoffquellen konnten 1,3-PD-tolerante, Rohglycerin-verwertende Stämme mit geringen Supplin-Anforderungen angereichert werden. Ein Scale-down der Anreicherungskultur in 96-Deep-Well-Platten gestattete die Untersuchung einer hohen Anzahl an Umweltproben. Als zweite Anreicherungsvariante mit hohem Durchsatz wurde die gemeinsame Inkubation mehrerer Umweltproben in einer Kultur (gruppierte Anreicherung) getestet. Eine Evaluierungsmethode für Isolate wurde entwickelt, um eine fundierte Stamm-Auswahl für die Fermentationsoptimierung zu treffen. Durch Kultivierung im Mikromaßstab konnte die Anzahl der Evaluierungsparameter stark erhöht werden. Die 1,3-PD-Produktion in Batchkultur ohne pH-Regelung wurde durch die Optimierung des pH-Puffersystems auf das dreifache gesteigert (max. 56 g/L 1,3-PD). 27 Stämme wurden unter anderem bezüglich Edukt- und Produkt-Toleranz evaluiert. 10 Stämme bildeten 1,3-PD bei hohen Produktvorlagen hinzu und erreichten so Endkonzentrationen von 80-110 g/L. 7 dieser 10 Isolate zeigten Wachstum und Produktbildung mit 200 g/L Rohglycerin und waren in der Produktion zum Teil gefördert im Vergleich zur Nutzung von reinem Glycerin. Die entwickelten Strategien konnten erfolgreich angewendet werden. Nach der Identifizierung ausgewählter Isolate wurden 2 Stämme der Art Clostridium butyricum, AKR17a und AKR102a, für die Fermentationsoptimierung im Bioreaktor ausgewählt.

The purpose of this work was the development of a custom-made screening strategy for the isolation of 1,3-propanediol high-performance strains. For this purpose an automatable high through-put procedure was to be developed. Based on a conventional batch enrichment of anaerobic, glycerol utilizing microorganisms from natural samples firstly through-put and selectivity were increased. By employing high concentrations of 1,3-PD and crude glycerol as well as different nutrient sources 1,3-PD tolerant and crude glycerol utilizing strains with low suppline demand could be enriched. A scale-down of the enrichment cultures in 96-deep-well plates allowed the analysis of a high number of natural samples. As a second enrichment procedure with a high through-put the combined incubation of several natural samples in one enrichment culture (grouped enrichment) was tested. An evaluation procedure for isolates was developed in order to make a well-founded strain selection for the optimization of fermentation. By cultivation on micro-scale the number of evaluation experiments could be notably increased. The 1,3-PD production for batch cultivation without pH regulation was increased threefold by optimization of the pH buffering system (max. 56 g/L 1,3-PD). 27 strains were evaluated, amongst other things, for educt and product tolerance. 10 strains produced additional 1,3-PD when high concentrations of the product had been added externally. Thus these 10 strains reached final concentrations in the range of 80 to 110 g/L and were further investigated. 7 of the 10 strains utilized 200 g/L crude glycerol for 1,3-PD production and several were stimulated in the 1,3-PD production compared to the utilization of pure glycerol. The here developed strategies were successfully applied. After the identification of selected isolates 2 strains of the species Clostridium butyricum, AKR17a and AKR102a, were chosen for further optimization of fermentation in a bioreactor system.
