Abstract

· Durch die Aufreinigung von Blattlausmaterial mittels Umkehrphasen-Hochleistungsflüssigkeitschromatographie (Reverse-Phase High Performance Liquid Chromatographie) in Kombination mit einem in vitro-Bioassay wurde für ein mutmaßliches Acyrthosiphon pisum-AKH die Aminosäure-Teilsequenz -NFST- erhalten. · Die Suche in den ersten bisher veröffentlichten Sequenzen des Genoms von A. pisum führte zu der Sequenz QVNFTPTWGQamid, die ein mutmaßliches adipokinetisches/ hypertrehalosämisches Peptid darstellt. · Der Stoffbestand der geflügelten (alaten) und der ungeflügelten (apteren) Morphe wurde im letzten Larven- und im Adultstadium analysiert. - Das Frischgewicht der ungeflügelten Morphe ist während des letzten Larvenstadiums und des Adultstadiums höher als das der geflügelten. Das Trockengewicht der Alatae ist nur am ersten Tag des Adultstadiums höher als das der Apterae. - Die geflügelten Blattläuse haben, abgesehen von Tag 10 des Adultstadiums, immer einen höheren Gehalt an Protein als die ungeflügelten. - Der Gehalt an Glykogen steigt bei beiden Morphen zum Ende des letzten Larvenstadiums stark an und nimmt dann mit steigendem Alter wieder ab. - Die geflügelte Morphe hat am Ende des letzten Larvenstadiums und während des Adultstadiums, außer an Tag 3, einen höheren Gehalt an freien Kohlenhydraten als die ungeflügelte Morphe. - Der Lipidgehalt ist bei den Alatae während des letzten Larvenstadiums und bis zu Tag 3 des Adultstadiums höher als bei den Apterae. Bei der geflügelten Morphe nimmt der Gehalt an Lipid bis zum 1. Tag des Adultstadiums zu und dann mit steigendem Alter wieder ab. - Protein ist in allen Altersstadien und bei beiden Morphen der vorherrschende Inhaltsstoff. Glykogen ist am Ende des letzten Larvenstadiums in größerer Menge vorhanden als freie Kohlenhydrate und Lipid, aber im späteren Adultstadium der quantitativ unbedeutenste Bestandteil. · Rosa gefärbte A. pisum haben ein höheres Frischgewicht als heller gefärbte. An Tag drei des Adultstadiums haben rosa gefärbte ungeflügelte Blattläuse einen höheren Kohlenhydratgehalt und rosa gefärbte geflügelte Tiere einen höheren Protein- und Glykogengehalt als heller gefärbte. · Der Einbau von Glucose und Trehalose in Glykogen ist bei den Adulttieren der geflügelten Morphe höher als bei den ungeflügelten Tieren. · Im Adultstadium baut die geflügelte Morphe mehr Acetat in Lipid ein als die ungeflügelte Morphe. Die Lipidsyntheserate steigt bei beiden Morphen von Tag 0 zu Tag 3 des Adultstadiums stark an. · Der Einbau von Acetat in die Lipidklassen Phospholipide, freie Fettsäuren, DAG und TAG konnte nachgewiesen werden. Die ungeflügelte Morphe baut einen höheren Anteil von Acetat in Phospholipide und weniger Acetat in TAG ein als die geflügelte. An Tag 3 des Adultstadiums synthetisieren die Apterae zusätzlich mehr freie Fettsäuren als die Alatae. Die geflügelte Morphe baut indessen an Tag 0 eine größere Menge an Acetat in DAG ein als die ungeflügelte Morphe und synthetisiert an Tag 3 weniger DAG und mehr TAG als zuvor an Tag 0. · Die Zugabe von 5 × 10-4 M Grybi-AKH führt zu einer tendenziellen Hemmung des Acetateinbaus in Gesamtlipid. · Methopren-behandelte geflügelte Tiere bauen mehr Acetat in TAG und weniger Acetat in freie Fettsäuren ein als die Kontrolltiere. · Aufgrund der Trehalase-Aktivität in der Hämolymphe wird in verdünnten Hämolymphproben, die bei Raumtemperatur gelagert werden, Trehalose zu Glucose abgebaut. · Die Hämolmyphe der adulten geflügelten Morphe hat einen höheren Trehalosegehalt als die der ungeflügelten. Der Gehalt an Trehalose nimmt bei den Alatae vom Larvenstadium zum Adultstadium zu. Die topikale Applikation von Grybi-AKH verändert die Trehalosekonzentration in der Hämolymphe nicht.

· Using Reverse-Phase High Performance Liquid Chromatographie in combination with an in vitro-bioassay a partial amino acid sequence for a putative Acyrthosiphon pisum-AKH (-NFST-) was obtained. · The search for AKH-like peptides in the previously published sequences of the genome of A. pisum revealed the sequence QVNFTPTWGQamid. · The body contents of organic substances of winged (alate) and unwinged (apterous) morphs of the last larval and adult stage was analysed. - The fresh weight of the unwinged morph is higher than the fresh weight of the winged morph during the last larval and adult stage. In adult aphids, the dry weight of the alatae is higher only at the first day. - Winged aphids have a higher protein content than the unwinged ones, except for 10 d-old adults. - In both morphs, the glycogen content increases at the end of the last larval stage and decreases again with increasing adult age. - The winged morph has a higher free carbohydrate content at the end of the last larval stage and during the adult stage, exept for 3 d-old adults, compared to the unwinged morph. - During the last larval stage until day 3 of the adult stage, the lipid content is higher in alatae than in apterae. In the winged morph the lipid content increases until the first day of the adult stage and decreases again with increasing age. - Protein constitutes the major portion of organic substances in all stages and in both morphs. In the last larval stage, more glycogen than free carbohydrate and lipid is present, but at the end of the adult stage the glycogen level drops below the levels of carbohydrate and lipid. · Pink coloured A. pisum have a higher fresh weight than pale coloured ones. 3 d-old pink coloured unwinged adults have a higher carbohydrate content and pink coloured winged adults a higher protein and glycogen content than pale coloured ones. · The incorporation of glucose and trehalose into glycogen is higher in winged aphids than in unwinged ones. · The incorporation of acetate into lipid is higher in the winged than in the unwinged morph. In both morphs, the rate of lipid synthesis increases from day 0 to day 3 of the adult stage. · The incorporation of acetate into the lipid classes phospholipids, free fatty acids, DAG, and TAG was demonstrated. The unwinged morph incorporates a greater proportion of acetat into phospholipid and a lower amount into TAG than the winged one. Additionally, on day 3 of the adult stage, the apterae synthesize more free fatty acids than the alatae. In contrast, on day 0 the winged morph incorporates a larger amount of acetate into DAG than the unwinged morph and on day 3 synthesises less DAG and more TAG than on day 0. · Addition of 5 × 10-4 M Grybi-AKH tends to inhibit the acetate-incorporation into total lipid. · Methoprene-treated winged aphids incorporate more acetate into TAG and less acetate into free fatty acids compared to the control animals. · Due to trehalase-activity in the hemolymph, trehalose is degraded to glucose when diluted haemolymph samples are stored at room temperature. · The adult winged morph has a higher haemolymph trehalose content than the unwinged one. The trehalose content increases in alatae from the larval to the adult stage. Topical application of Grybi-AKH does not alter the trehalose concentration in the haemolymph.
