Abstract

Im Rahmen dieser Arbeit sollten Beiträge zu einem neuen synthetischen Zugang zur Substanzklasse der polycyclischen Tetramsäure-Makrolactame (PTM) geleistet werden. Dafür sollte ein vereinfachtes, ursprünglich von trans-Discodermid abgeleitetes, Modell hergestellt werden. Dabei wurde auf der vorrangegangenen Arbeit von Dr. Markus Winterer aufgebaut. Der Grundgedanke zur Synthese des Modells war eine Ketenylidentriphenylphosphoran vermittelte Kupplung eines Modellaldehyds und einer Ornithin-Tetramsäure. Die von Winterer aufgestellte Synthese für den Modellaldehyd wurde optimiert und ihre Gesamtausbeute von ehemals 8% auf 39% über sieben Stufen gesteigert. Die Synthese des Bisylids aus einer Ornithin-abgeleiteten Tetramsäure und Ketenylidentriphenylphosphoran verlief zwar erfolgreich, die Verbindung erwies sich jedoch als völlig unreaktiv. Die Begründung wurde in der Bildung eines internen Salzes gesucht. Versuche das Bisylid zu aktivieren scheiterten. Das Modell sollte alternativ über ein Amidylid-Salz aufgebaut werden. Der 9 BBN-Komplex von L-Ornithin sollte mit Ketenylidentriphenylphosphoran zum korrespondierenden Salz umgesetzt und dieses mit dem Modellaldehyd zum entsprechenden Kupplungsprodukt reagieren. Diese Reaktion lieferte neben schlechten Ausbeuten auch schlechte cis:trans-Verhältnisse und anhand der Kopplungskonstanten den Beweis, dass der Naturstoff an dieser Stelle nicht, wie von Winterer angenommen, trans-konfiguriert ist. Aufgrund dieses Befundes wurde die Synthese durch Austausch des kumulierten Ylids gegen ein Phosphonat und entsprechende Modifizierung des Modellaldehyds angepasst. Der Modellaldehyd wurde erfolgreich zur Säure umgesetzt und diese mit dem zuvor synthetisierten 9 BBN-Komplex zum Kupplungsprodukt verknüpft. Dieser Komplex erwies sich jedoch als unerwartet stabil, sodass er sich nicht entschützen ließ. Alternativ wurde kommerziell erhältliches geschütztes Ornithin erfolgreich verestert und in Anwesenheit von TFA hydriert, um einen Ringschluss zu unterdrücken. Das korrespondierende Salz konnte mit dem Modellaldehyd erfolgreich verknüpft und der Methylester zu verseift werden. Die Einführung der Tetramsäurefunktion war erfolgreich, ebenso die 3 Acylierung mit Ketenylidentriphenylphosphoran. Die folgende Entschützung der Aldehydfunktion gestaltete sich als äußerst schwierig und konnte letztendlich nur in konzentrierter TFA erfolgreich durchgeführt werden. Alle Versuche den Ringschluss zum fertigen Modell zu erreichen scheiterten. Es wurde gemutmaßt, dass es sich um ein sterisches Problem handeln könnte und das System nicht in die richtige Konformation findet, um die Reaktion einzugehen. Um dem System eine andere Umgebung zu geben, wurde die Reaktion an der Festphase versucht, doch auch hier lieferte sie nicht das gewünschte Produkt. Somit kann diese Route nicht als zielführend angesehen werden. Weitere Studien des komplexen Systems sind nötig. Hinsichtlich der Tatsache, dass viele PTM, darunter auch Discodermid, ß Hydroxyornithin enthalten, wurde dieses nebenbei hergestellt. Ausgehend von Aminopropanol konnte das voll geschützte ß-Hydroxyornithin erfolgreich mit einer Gesamtausbeute von 12% synthetisiert werden.

This thesis should contribute to a new synthetic access to the class of polycyclic tetramate macrolactams (PTM). A simplified model, derived from trans-discodermide, was to be synthesized. The work was based upon the preceding work of Dr. Markus Winterer. The basic idea for the synthesis was the ketenylidene triphenylphosphorane mediated coupling of a model aldehyde and a tetramic acid. The synthesis for the model aldehyde by Winterer was optimized and the overall yield increased from formerly 8% to 39% over seven steps. The synthesis of the desired bisylide was successful but the compound turned out to be totally unreactive. The reason could be the formation of an internal salt. Attempts to activate the bisylide failed. It was attempted to synthesize the model via an amide ylide salt. The 9-BBN-complex of L-ornithine should react with ketenylidene triphenylphosphorane to form the corresponding amide ylide salt and react with the model aldehyde to yield the coupling product. The salt could be obtained and coupled with the model aldehyde successfully. The reaction resulted in bad yields as well as bad cis:trans ratios and the proof that the natural product is not trans-configurated as supposed by Winterer. Thus, the synthesis was adapted by exchanging the cumulated ylide with a phosphonate as well as the appropriate modification of the model aldehyde. The aldehyde was successfully converted to the acid and coupled with the complex to yield the product. The 9-BBN-complex turned out to be unexpectedly stable and could not be cleaved. Therefore, the coupling product should be obtained from commercially available protected ornithine which was converted into the methyl ester and hydrogenated in the presence of TFA to avoid ring formation. The resulting salt could be coupled with the model aldehyde and the present methyl ester was saponified to the acid. The introduction of the tetramic acid moiety was successful, as well as the 3-acylation with ketenylidene triphenylphosphorane. The following deprotection of the aldehyde turned out to be difficult and could only be achieved with concentrated TFA. All attempts to achieve the ring closure to the target molecule failed. It was supposed, that it could be because of sterically reasons that the system does not find to the right conformation to react. To offer the system a different surrounding, the reaction was attempted on the solid phase, but it also did not lead to the desired product. Therefore, this route cannot be regarded as productive. Further investigation is needed. Regarding the fact that many PTM, including discodermide, contain ß-hydroxy ornithine, this was also synthesized. Starting from aminopropanol the fully protected ß hydroxy ornithine could be synthesized with an overall yield of 12%.
