Xanthohumol is the major prenylated chalcone found in hops, and it has been shown to exhibit various biological effects. However, xanthohumol effects on liver cells or in liver diseases, respectively, are widely unknown. In the present work, first the effects of xanthohumol on hepatic stellate cells (HSC), the central mediators of liver fibrogenesis, were analyzed. Xanthohumol inhibited the activation of primary human HSC and induces apoptosis in activated HSC in vitro in a dose dependent manner (0-20 µM). In contrast, xanthohumol doses as high as 100 µM did not impair viability of primary human hepatocytes (PHH). However, in both cell types xanthohumol inhibited NF-kappaB activation and expression of NF-kappaB dependent proinflammatory chemokines. In vivo, feeding of xanthohumol reduced levels of serum transaminases and hepatic expression of proinflammatory genes in a murine model of non-alcoholic steatohepatitis (NASH). Moreover, xanthohumol treatment significantly inhibited hepatic expression of profibrogenic genes and activation of HSC in vivo. Next, xanthohumol effects on hepatocellular carcinoma (HCC) cells were investigated. Xanthohumol concentration of 25 µM induced apoptosis in two HCC cell lines (HepG2 and Huh7). Furthermore, xanthohumol repressed proliferation and migration, as well as TNF induced activation of the transcription factor NF-kappaB and interleukin-8 expression in both cell lines at even lower concentrations. Finally, to evaluate the safety profile of xanthohumol female BALB/c mice were fed with a xanthohumol enriched diet for three weeks, achieving a daily dose of approximately 1000 mg xanthohumol /kg body weight per day. Macroscopical and histopathological examination of liver, kidney, colon, lung, heart, spleen and thymus revealed no signs of xanthohumol-toxicity, and biochemical serum analysis confirmed normal organ function. Further, serum glucose levels and hepatic glycogen content as well as hepatic CYP2E1 mRNA expression levels were unaffected by xanthohumol treatment. In addition, also mRNA expression of several genes indicative of early hepatic inflammation and fibrosis, a hallmark of chronic liver injury, did not differ between xanthohumol treated and control mice. In conclusion, xanthohumol has the potential to ameliorate NASH induced liver injury as well as different pro-tumorigenic mechanisms known to promote HCC progression. Together with the good safety profile these data suggest the potential use of xanthohumol as a functional nutrient or therapeutic agent to prevent or treat chronic liver diseases like NASH or HCC.

Für Xanthohumol, ein im Hopfen vorkommendes prenyliertes Chalcon, konnte eine Vielzahl von biologischen Effekten nachgewiesen werden. Wirkungen von Xanthohumol auf Leberzellen oder bei Lebererkrankungen sind jedoch weitgehend unbekannt. In dieser Arbeit wurden zunächst Effekte von Xanthohumol auf hepatische Sternzellen (HSC), den zentralen Mediatoren der Leberfibrogenese, analysiert. Xanthohumol konnte dosisabhängig (0-20 µM) die Aktivierung von primären humanen HSC hemmen sowie Apoptose in aktivierten HSC induzieren. Im Gegensatz dazu wurde die Viabilität von primären humanen Hepatozyten (PHH) selbst durch hohe Dosen von Xanthohumol (100 µM) nicht beeinträchtigt. In beiden Zelltypen hemmte Xanthohumol jedoch sowohl die NF kappaB Aktivität als auch die Expression von NF kappaB abhängigen proinflammatorischen Chemokinen. In vivo konnte durch Zufütterung von Xanthohumol in einem murinen Fütterungsmodell zur Induktion einer Nichtalkoholischen Fettleberhepatitis (NASH) eine Erniedrigung der Serumtransaminasespiegel und eine Hemmung der Expression von proinflammatorischen Genen erreicht werden. Außerdem verminderte die Behandlung mit Xanthohumol signifikant die hepatische Expression von profibrogenen Genen sowie die Aktivierung von HSC in vivo. Als nächstes wurden die Effekte von Xanthohumol auf hepatozelluläre Karzinomzellen (HCC) untersucht. Mit einer Xanthohumolkonzentration von 25 µM konnte in zwei HCC Zelllinien (HepG2 und Huh7) Apoptose induziert werden, wohingegen bereits deutlich geringere Konzentrationen sowohl Proliferation, Migration als auch TNF vermittelte Aktivierung des Transkriptionsfaktors NF kappaB sowie Expression von Interleukin-8 hemmten. Um das Sicherheitsprofil von Xanthohumol zu bestimmen, wurden weibliche BALB/c Mäuse drei Wochen lang mit einer Xanthohumol-angereicherten Diät gefüttert. Eine tägliche Xanthohumolaufnahme von ca. 1000 mg pro kg Körpergewicht wurde ermittelt. Makroskopische und histopathologische Untersuchung von Leber, Niere, Kolon, Lunge, Herz, Milz und Thymus zeigten keine Anzeichen einer durch Xanthohumol vermittelten Toxizität. Biochemische Serumanalysen wiesen auf normale Organfunktion hin. Die Behandlung mit Xanthohumol hatte ferner keine Auswirkung auf den Blutzuckerspiegel, den hepatischen Glykogengehalt sowie auf die CYP2E1 mRNA Expression in der Leber. Weiterhin zeigte sich zwischen Xanthohumol-behandelten und unbehandelten Mäusen kein signifikanter Unterschied in der Expression von Genen, die mit Leberentzündung und Leberfibrose, eine Folge chronischer Leberschädigung, in Verbindung gebracht werden. Zusammenfassend konnte in dieser Arbeit gezeigt werden, dass Xanthohumol sowohl das Potenzial zur Behandlung einer NASH-induzierten Leberschädigung als auch zur Hemmung von verschiedenen pro kanzerogenen Mechanismen, die eine HCC Progression begünstigen, besitzt. In Anbetracht des guten Sicherheitsprofils von Xanthohumol lässt sich somit seine Verwendung als Nahrungsergänzungsmittel oder als Arzneimittel zur Vorbeugung oder Behandlung von chronischen Lebererkrankungen wie NASH oder HCC vorstellen.
