Abstract

In order to reveal the influence of substituents at the bridging position of bis(pyrazol-1-yl)acetato ligands, the coordination behavior the model ligand 2,2-bis(3,5-dimethylpyrazol-1-yl)propanoic acid (L1) was investigated in the first part of this thesis. It was observed that the introduced methyl group has a crucial effect on the structure and reactivity of transition metal complexes of ligand L1. Steric interactions of the methyl group at the bridging position and the pyrazolyl substituents at position 5 cause a twist of one pyrazolyl unit. The steric tension in the bisphosphine complex [Ru(L1)Cl(PPh3)2] is responsible for the enhanced dissociation of one PPh3 ligand. The resulting coordinatively unsaturated 16 VE fragment [Ru(L1)Cl(PPh3)] readily reacts with N2 molecules of the inert gas atmosphere to form the dinitrogen complex [Ru(L1)Cl(N2)(PPh3)] which represents the first example for a N2 complex bearing a bis(pyrazol-1-yl)acetato ligand. In contrast, treatment of [Ru(L2)Cl(PPh3)2] (L2 = 2,2-bis(3,5-dimethylpyrazol-1-yl)acetic acid) with N2 showed only traces of a N2 product in the IR spectrum. Unsaturated fragments such as [Ru(L1)Cl(PPh3)] are important intermediates in organometallic and inorganic transformations and might be of interest as potential catalysts and for the activation of small molecules. In the second part of this thesis, the new N,N,O ligand 2,2-bis(3,5-dimethyl-4-vinylpyrazol-1-yl)acetic acid (L3) was synthesized in four steps. The advantage of this ligand is that the functional units for polymerization are no longer positioned at the bridging carbon atom and thus do not influence the remaining space of κ3-N,N,O bound transition metal fragments. Ligand L3 was copolymerized with either methyl methacrylate (MMA) or ethylene glycol dimethacrylate (EGDMA) and the resulting solid phases have been treated with appropriate precursor compounds to form incorporated tricarbonyl complexes [M(L3)(CO)3] (M = Mn, Re). The successful incorporation of the metals was verified by ICP-AES and AAS measurements. IR spectroscopic investigations on these polymers revealed that the embedded metal centers are bound in a fac-κ3-N,N,O coordination mode. Furthermore, an embedded copper(II) complex was synthesized by heterogeneous reaction of a MMA copolymer of ligand L3 with CuCl2. UV/Vis spectroscopic investigations revealed a bathochromic shift by 64 nm compared to the UV/Vis spectrum of the homogeneous complex [Cu(L3)2]. This implies that the copper(II) centers embedded in this copolymer are mainly coordinated by one ligand moiety and the unwanted formation of bisligand complexes [M(L)2] can be prevented. In the third part of this work, ligand L3 was used for the generation of imprinted polymers. For this purpose, two template complexes [Ru(L3)(BF)(PPh3)] (BF = benzoylformate) and [Ru(L3)(NOG)(PPh3)] (NOG = N-oxalylglycine) have been synthesized. Since NOG is isostructural to 2-oxoglutaric acid, [Ru(L3)(NOG)(PPh3)] can be considered as a model complex for a 2-OG-dependent iron(II) oxygenase inhibited by NOG. Thus, it is well suited as a “dummy” complex for the generation of an imprinted polymer which might mimic the catalytic activity of such an oxygenase enzyme. Both template complexes have been copolymerized with EGDMA in the presence of a porogene solvent. The resulting solid phases have been analyzed by UV/Vis spectroscopy and it was found that the absorption maxima of the polymerized complexes are almost identical to those recorded for the corresponding homogeneous complexes which proves the successful incorporation. The imprints have been generated by treating the polymers with excess of PMe3. ICP-AES measurements have shown that up to 51% of the ruthenium templates can be removed by this procedure. In upcoming studies, it has to be examined whether these imprinted polymers are able to catalytically oxidize substrates such as cyclohexene on addition of iron(II) and 2-OG in presence of dioxygen or a oxidizing agent such as H2O2. First experiments of incorporation of the generated cavities with iron(II) are on their way. In a sub project of this thesis it was found that the complexes [Ru(L1)Cl(CO)2] and [Ru(L2)Cl(CO)2] are able to selectively catalyze the epoxidation of cyclohexene with iodosyl benzene. During this theis, a TON of up to 20.6 was obtained for [Ru(L2)Cl(CO)2] by tuning the reactant/catalyst ratio. In a second sub-project, the small molecule inhibitor 2-(1-chloro-4-hydroxyisoquinoline-3-carboxamido)acetic acid was synthesized according to literature procedures and provided to the group of Prof. Dr. K.-U. Eckardt (Medicinal Clinic 4, Nephrology and Hypertension, Friedrich-Alexander-University Erlangen-Nürnberg). It was found that this compound is able to stabilize HIF (hypoxia-inducible factor) and induce HIF target genes in vitro and in vivo. Preconditional HIF activation by inhibitors might be used for the treatment of ischemic diseases or for organ preservation in transplantation.

Abstract

Im ersten Teil der Arbeit wurde das Koordinationsverhalten des neuen N,N,O-Liganden 2,2-Bis(3,5-dimethylpyrazol-1-yl)propansäure (L1) untersucht. Sowohl die Struktur als auch die Reaktivität von Komplexverbindungen dieses Liganden werden stark durch die Methylgruppe an der Brückenposition beeinflusst. Sterische Wechselwirkungen zwischen den Methylgruppen führen zu einer Verdrehung eines der beiden Pyrazoldonoren. Im Bisphosphin-Komplex [Ru(L1)Cl(PPh3)2] hatte dies die Verdrängung eines PPh3-Liganden und somit die Ausbildung eines koordinativ ungesättigen 16VE-Fragmentes [Ru(L1)Cl(PPh3)] zur Folge. Durch Reaktion mit vorhandenen N2-Molekülen des Schutzgases entstand der Distickstoff-Komplex [Ru(L1)Cl(N2)(PPh3)]. Diese Verbindung ist das erste bekannte Beispiel für einen Distickstoff-Komplex eines Bis(pyrazol-1-yl)essigsäure Liganden und ist im Hinblick auf die Aktivierung kleiner Moleküle von besonderem Interesse. Blindversuche mit dem Liganden 2,2-Bis(3,5-dimethylpyrazol-1-yl)essigsäure (L2) zeigten, dass dieser nicht zur Bildung eines analogen Distickstoff-Komplexes fähig ist. Im zweiten Teil der Arbeit wurde der zur Copolymerisation geeignete N,N,O-Ligand 2,2-Bis(3,5-dimethyl-4-vinyl-pyrazol-1-yl)essigsäure (L3) dargestellt. Der Vorteil dieses neuartigen Liganden ist, dass sich die polymerisationsaktiven Substituenten nicht wie bisher üblich am Brückenkohlenstoffatom befinden, sondern als Substituenten in Position 4 der Pyrazolringe angebracht sind und somit das Koordinationsverhalten des Liganden nicht nachteilig beeinflussen. Durch Copolymerisation mit Methylmethacrylat (MMA) oder Ethylenglykoldimethacrylat (EGDMA) erzeugte Festphasen wurden mit geeigneten Metallprecursor-Verbindungen zu immobilisierten Komplexen [M(L3)(CO)3] (M = Mn, Re) umgesetzt. Mittels AAS und ICP-AES Messungen konnte die Aufnahme der Übergangsmetalle in die polymeren Festphasen bestätigt werden. Durch IR-Spektroskopie konnte belegt werden, dass die im Polymer verankerten Ligand-Fragmente in der Lage sind Metallzentren mit einem fac-κ3-N,N,O Motiv zu binden. Weiterhin wurde ein MMA-Copolymer des Liganden L3 in einer heterogenen Reaktion mit CuCl2 umgesetzt. Das UV/Vis-Spektrum des resultierenden kupferhaltigen Polymers zeigt eine bathochrome Verschiebung des Absorpionsmaximums um 64 nm im Vergleich zum homogenen Bisligand-Komplexes [Cu(L3)2]. Aufgrund der Quervernetzung in der polymeren Festphase kann daher die Bildung von unerwünschten Komplexen des Typs [M(L)2] umgangen werden. Im weiteren Verlauf wurde der Ligand L3 zur Erzeugung molekular geprägter Polymere genutzt. Die synthetisierten Templatkomplexe [Ru(L3)(BF)(PPh3)] (BF = Benzoylformiat) und [Ru(L3)(NOG)(PPh3)] (NOG = N-Oxalylglycin) können als strukturelle Modelle für die aktiven Zentren 2‑Oxoglutarat-abhängiger Eisen(II)-Enzyme betrachtet werden. Hervorzuheben ist, dass N‑Oxalylglycin strukturanalog zum natürlich vorkommenden Cosubstrat 2-Oxoglutarsäure ist und somit einen Inhibitor für 2-OG-abhängige Eisen(II)-Oxygenasen darstellt. Daher eignet sich [Ru(L3)(NOG)(PPh3)] besonders gut als Templat für die Erzeugung eines geprägten Polymers, mit dem die katalytische Funktion solcher Enzyme nachgeahmt werden kann. Die Templatkomplexe wurden in Gegenwart von EGDMA und einem porogenen Solvens copolymerisiert. Durch UV/Vis‑Spektroskopie konnte die erfolgreiche Einbindung der Komplexe in eine Polymermatrix belegt werden. Um die erwünschte Prägung zu erzeugen, wurden die Polymere mit PMe3 behandelt. Durch ICP-AES Messungen konnte ein deutlicher Rückgang des Rutheniumgehaltes von bis zu 51 % beobachtet werden. Zukünftige Studien werden zeigen, ob diese geprägten Umgebungen bei Zugabe von Eisen(II) und 2‑Oxoglutarat dazu fähig sind, Substrate wie beispielsweise Cyclohexen mit atmosphärischem Sauerstoff oder einem Oxidationsmittel wie H2O2 katalytisch zu oxidieren. In ersten Versuchen hierzu wurden die geprägten Umgebungen bereits mit Eisen(II) besetzt. In einem Teilprojekt dieser Arbeit konnte gezeigt werden, dass die Komplexe [Ru(L1)Cl(CO)2] und [Ru(L2)Cl(CO)2] die selektive Epoxidierung von Cyclohexen zu Cyclohexenoxid katalysieren. Als Oxidationsmittel wurde Iodosobenzol eingesetzt. Für den Komplex [Ru(L2)Cl(CO)2] wurde im Rahmen dieser Arbeit eine Wechselzahl von 20.6 ermittelt. In einem zweiten Teilprojekt, wurde der Prolylhydroxylase-Inhibitor N-((1-Chlor-4-hydroxyisochinolin-3-yl)carbonyl)glycin nach Literaturvorschriften synthetisiert und an die AG Prof. Dr. K.‑U. Eckardt (Medizinische Klinik 4, Nephrologie und Hypertensiologie, Friedrich-Alexander-Universität Erlangen-Nürnberg) für Experimente zur Stabilisierung von HIF (Hypoxie-induzierter Faktor) bereitgestellt. Es zeigte sich, dass die Verbindung in der Lage ist, HIF 1α zu stabilisieren und HIF-Zielgene sowohl in vitro als auch in vivo zu induzieren. Die Aktivierung von HIF durch Inhibitormoleküle kann zur Behandlung von ischämischen Erkrankungen oder die Konservierung von Spenderorganen bei Transplantationen hilfreich sein.
