Sustainable bio- and organocatalytic syntheses of enantiomerically pure beta-amino acids and derivatives

Abstract

The aim of this thesis was the development of novel sustainable enantioselective synthetic pathways towards enantiomerically highly enriched beta-amino acids and derivatives, in particular short-chain aliphatic beta-3-amino acids and derivatives, which are an important product class for pharmaceutical applications. During the development particular attention was paid to sustainability aspects through realization of green chemistry principles by incorporation of bio- and organocatalytic reactions and modern environmental assessment tools (EATOS and Umberto/Sabento). Chapter 1 presented selected examples for the application of beta-amino acids as chiral building blocks in the synthesis of pharmaceutically active compounds (API´s). Subsequently the aims of this work were defined. Chapter 2 described novel approaches towards novel asymmetric bicyclic amidine and phase-transfer catalyzed aza-Michael reactions. The potential of the concept based on the use of the bicyclic amidine (R)-31 catalyst in terms of a general synthetic pathway towards short-chain beta-amino acids is rather limited, since the observed reactivity and enantioselectivity in the aza-Michael was very low (conversion <25%, enantioselectivity <5% ee). Additionally, a novel concept using ammonium (cinchonidinium salts) and phosphonium catalysts (P-chirogenic catalysts were gratefully supported by Prof. S. Jugé, University of Burgundy) for the aza-Michael reaction of benzoyl carbamate nucleophiles to activated alpha,beta-unsaturated acceptors was investigated. Even though the described protocol does not yet meet the criteria of a general synthetic platform for the access to the desired target molecules, the proof of concept for the first asymmetric intermolecular phase-transfer catalyzed aza-Michael reaction was given. The best result (81% yield and 22% ee) was achieved using 30 mol% of the Maruoka catalyst 46. Chapter 3 described the successful development of a novel sustainable solvent-free chemoenzymatic process for the synthesis of various aliphatic short-chain beta-3-amino acid esters 22 in moderate to satisfying yields (19-36%) and with high to excellent enantiomeric excess in the range of 93-99% ee. In spite of the high enantiomeric excess for the esters 22, the corresponding amides 80 were obtained in less satisfactory enantiomeric excesses in the range of 36-64% ee (although good yields were obtained). The enantiomeric ratio E of the Candida antarctica lipase B catalyzed resolution step (via aminolysis) was analyzed for the synthesis of various aliphatic short-chain beta-3-amino esters 22. The enantiomeric ratio for the standard substrate ethyl rac-(3-benzylamino)butanoate (rac-22a) was determined to E= 27. After analysis of the enantiomeric ratio E of the substrate scope, the influence of the ester residue and the substitution pattern at the aromatic moiety of the nitrogen donor were studied. Accordingly a slightly improved enantiomeric ratio E was observed by elongation of the ester residue to n-propyl instead of ethyl (E= 33). Variation of the substitution pattern at the aromatic moiety of the nitrogen donor led to an enhanced enantiomeric ratio of E= 37 (methyl group in 4-position at the aromatic ring). Subsequently the process development was focused based on ecological considerations and green chemistry principles. An improved process that overcomes the limitations of the standard process (based on solvent consuming column chromatography and separate unit-operations) was developed. Key achievements of this greener alternative include significant reduction of organic solvent waste (through replacement of column chromatography), replacement of chemicals by greener alternatives (e.g. substitution of Pd(OH)2/C by Pd/C), direct mass transfer to the next unit operation without the need of isolating an intermediate and recycling of the biocatalyst. After establishment of the batch process, the feasibility of a technical scale-up (continuous production) was studied at the Institute of Technical Biocatalysis in Hamburg (cooperation with the working group of Prof. Dr. A. Liese). The results demonstrated that continuous operation is possible in principle (using a tube reactor and a packed bed reactor with immobilized enzyme). A high space-time yield of STY ~75 gh-1L-1 was achieved, whereas the crude product ethyl (S)-(3-benzylamino)butanoate ((S)-22a) was yielded with high enantiomeric excess (95% ee). Finally the synthesized enantiomerically highly enriched (S)-3-aminobutanoic acid ((S)-75a, 99% ee) was successfully utilized as a building block within syntheses towards derivatives 83d-f of the active pharmaceutical ingredient (API) Levomepromazine ((R)-83c). The receptor binding studies of the Levomepromazine derivatives 83b, rac-83c, rac-83d and (S)-83d revealed a relationship between the position and the absolute configuration of the stereogenic center and the affinity towards the examined receptors (assay carried out by research group of Prof. P. Gmeiner, Department of Chemistry and Pharmacy, University Erlangen-Nuremberg). Chapter 4 described the environmental assessment of two alternative synthesis of S)-3-aminobutanoic acid hydrochloride ((S)-75a xHCl) via mass balancing and SHE (safety, health and environment) assessment with the software programs EATOS and Umberto/Sabento. The software tools were used at the process development stage to identify optimization potential within the standard laboratory procedure. After identification of the weaknesses the environmental assessment methods were used to support the decision process during the process development of more sustainable process alternatives. The analysis of key environmental indicators such as, e.g., S-1, E-factor and PEI, revealed a significantly improved sustainability of the optimized process in comparison to the original process. A reduction of the E-factor by more than eight times was achieved in addition to a reduction of the PEI (total) by more than 60 times. This fact also indicates a significant diminishment of hazardous substances for the optimized process.

Abstract

Das Ziel der vorliegenden Arbeit war die Entwicklung neuer enantioselektiver synthetischer Zugänge zu beta-Aminosäuren und deren Derivaten, im speziellen der Verbindungsklasse der kurzkettigen, aliphatischen enantiomerhochangereicherten beta-3-Aminosäuren und deren Derivaten, die insbesondere als chirale Bausteine im Bereich der Synthese von Pharmazeutika Anwendung finden. Während dieses Entwicklungsprozesses wurde der Berücksichtigung von Nachhaltigkeits-aspekten, wie den Prinzipien der grünen Chemie und der Verwendung von bio- und organokatalytischen Verfahrensweisen besondere Aufmerksamkeit geschenkt. Dazu wurden insbesondere moderne Softwareprogramme wie EATOS und Umberto/Sabento zur Bewertung der Nachhaltigkeit der Synthesen verwendet. In Kapitel 1 wurden einige ausgewählte Beispiele, in denen beta-Aminosäuren als chirale Bausteine zur Herstellung von pharmazeutisch aktiven Verbindungen Verwendung finden präsentiert. Anschließend wurden die Zielsetzungen dieser Arbeit definiert. In Kapitel 2 wurden neuartige asymmetrische aza-Michael Reaktionen, in denen bizyklische Amidine bzw. Phasentransfer-Katalysatoren als Organokatalysatoren Verwendung finden untersucht. Aufgrund der experimentell beobachteten geringen Reaktivität und Enantioselektivität (Umsatz <25%, <5% ee) stellt das Verfahren, welches den bizyklischen Organokatalysator (R)-31 verwendet keinen generellen Zugangsweg zu der gewünschten Verbindungsklasse der aliphatischen kurzkettigen beta-3-Aminosäuren dar. Weiterhin wurde ein neuartiges Konzept untersucht, bei dem Ammonium- (Cinchonidinium Salze) und Phosphonium-Katalysatoren (die Phosphor chiralen Katalysatoren wurden von Prof. S. Jugé, Universität Burgund, zur Verfügung gestellt) zur Addition von Carbamate-Nukleophilen an alpha,beta-ungesättigte Akzeptoren eingesetzt werden. Obwohl das beschriebene Verfahren noch nicht den Kriterien einer generellen Syntheseplattform entspricht, konnte dennoch der „proof of concept“ für die erste intermolekulare asymmetrische Phasen-transfer katalysierte aza-Michael Reaktion erbracht werden. Das beste Ergebnis wurde dabei unter Verwendung von 30 mol% des Maruoka-Katalysators 46 erzielt (81% Ausbeute und Enantiomerenüberschuss von 22% ee. In Kapitel 3 wurde die erfolgreiche Entwicklung eines chemoenzymatischen Eintopf-Verfahrens beschrieben, mit dem eine Reihe unterschiedlich substituierter aliphatischer kurzkettiger beta-3-Aminosäureester in moderaten Ausbeuten (19-36%) und mit hohen bis exzellenten Enantiomerenüberschüssen (93-99% ee) synthetisiert werden können. Die entsprechenden Amide 80 wurden im Gegensatz zu den hohen Enantiomerenüberschüssen der Ester 22 in geringeren Enantiomerenüberschüssen im Bereich von 36-64% ee erhalten, wobei die erzielten Ausbeuten in einem guten Bereich lagen (46-65% isolierte Ausbeute). Der Selektivitätswert E der Candida antarctica Lipase B katalysierten Racematspaltung (via Aminolyse) wurde separat für die Herstellung einer Reihe von beta-3-Aminosäureestern 22 untersucht. Die Enantioselektivität für das Standardsubstrat Ethyl rac-(3-benzylamino)butanoat (rac-22a) wurde mit E= 27 bestimmt. Nach der Untersuchung des Selektivitätswertes E der Substratbreite wurde der Einfluss des Substitutionsmusters des Esterrestes eingehend untersucht. Bei Verlängerung der Esterkette zu n-Propyl anstelle von Ethyl eine Selektivitätssteigerung auf E= 33 beobachtet werden. Durch Variation des Substitutionsmusters des aromatischen Ringes am Stickstoffdonor (Methyl- bzw. Methoxy-Substituenten an der 2-, 3-, bzw. 4-Position) konnte Selektivitätswertes E bezüglich des 4-Methyl substituierten Substrates auf E= 37 gegenüber dem Standardsubstrat gesteigert werden. Im Anschluss daran wurde die Prozessentwicklung zum gewünschten Endprodukt, der freien beta-Aminosäure (S)-74a) fokussiert, wobei speziell ökologische Überlegungen auf Basis der zwölf Prinzipien der grünen Chemie berücksichtigt wurden. Ausgehend von dem Standard-Verfahren wurde ein verbessertes Verfahren entwickelt, dass die vorherigen Limitierungen (säulenchromatographische Aufreinigung, Isolierung und Aufarbeitung nach der Reaktion etc.) überwindet. Die Haupterfolge bestanden dabei in der Abfallreduktion durch Substitution der säulenchromatographischen Aufreinigung, dem Ersatz von Chemikalien (z.B.: Substitution von Pd(OH)2/C durch Pd/C) durch weniger toxischen Alternativen, direktem Massentransfer von einem zum nächsten Prozessschritt ohne notwendige Isolierung von Intermediaten und Recycling des Biokatalysators. Nach der Etablierung des Batch-Verfahrens wurde die Möglichkeit der Eignung bzgl. technischer Aufskalierung (kontinuierliche Verfahrensführung) am Institut für Technische Biokatalyse in Hamburg untersucht (Kooperation mit der Arbeitsgruppe von Prof. Dr. A. Liese). Die Ergebnisse zeigten, dass die kontinuierliche Prozessführung (unter Verwendung eines Schlauchreaktors und eines mit immobilisiertem Enzym beladenden Festbettreaktors) prinzipiell möglich ist. Eine hohe Raumzeitausbeute (für den Festbett-Enzymreaktor) im Bereich von ~75 gh-1L-1 wurde erreicht, wobei das Rohprodukt (S)-(3-Benzyl-amino)butansäureethylester ((S)-22a) in hoher Enantiomerenreinheit von 95% ee erhalten wurde. Abschließend wurde das zuvor synthetisierte hoch enantiomerenangereicherte Produkt S)-3-Aminobutansäure ((S)-75a, 99% ee), als Baustein zur Synthesen der Pharmaderivate 83d-f von R-Levomepromazin ((R)-83c) eingesetzt Die Rezeptorbindungstests der Levomepromazinderivate 83b, rac-83c, rac-83d und (S)-83d offenbarten eine Korrelation zwischen der Position und absoluten Konfiguration des Stereozentrums und der Affinität gegenüber den untersuchten Rezeptoren (der Assay wurde von der Arbeitsgruppe von Prof. Dr. P. Gmeiner, Lehrstuhl für Chemie und Pharmazie, Universität Erlangen-Nürnberg) In Kapitel 4 wurde die ökologische Bewertung zweier alternativer Syntheserouten von (S)-3-Aminobutansäurehydrochlorid ((S)-75a xHCl), mittels Massenbilanzierung und SHE-Gewichtung (safety, health and environment – Sicherheit, Gesundheit und Umwelt) unter Verwendung der Software Programme EATOS und Umberto/Sabento durchgeführt. Dabei wurden die Software Programme EATOS und Umberto/Sabento bereits während der Prozessentwicklung verwendet, um das Optimierungspotential des Standardverfahrens zu analysieren. Nach der Identifikation von Prozesschwachstellen wurde die ökologische Bewertung genutzt um den Entscheidungsprozess während der Prozessentwicklung in Richtung einer Nachhaltigkeitsoptimierung zu unterstützen. Die Analyse der Schlüsselindizes wie Selektivität S-1, E-Faktor und PEI ergaben eine signifikante Verbesserung in Bezug auf die Nachhaltigkeit des optimierten Verfahrens. Der E-Faktor wurde um einen Faktor von über acht reduziert, wobei der potentielle Umwelteinfluss sogar um einen Faktor von mehr als 60 reduziert werden konnte. Insgesamt belegen diese Kennzahlen eine erhebliche Reduzierung des Gefahrenpotentials für den optimierten Prozess.
