Abstract

Saccharomyces cerevisiae cells control their cell size at a point in late G1 called Start. This work describes a negative role for the Sin3/Rpd3 histone deacetylase complex in the regulation of cell size at Start. Yeast cells must grow to a critical cell size, before they can enter S-Phase, and commit to a new cell cycle. In response to cellsize, the Cln3/Cdc28 kinase activates in late G1 the SBF-dependent transcription of G1-Cyclins. This work shows, that initiation of G1/S-specific transcription of CLN1, CLN2 and PCL1 in a sin3 strain and a rpd3 strain occurs at a reduced cell size compared with a wild-type strain. In addition, inactivation of the transcriptional regulator SIN3 partially suppressed a large cln3 mutant, causing sin3cln3 double mutants to start the cell cycle at wildtype size. Chromatin immunoprecipitation results demonstrate, that Sin3 and Rpd3 are recruited to promoters of SBF (Swi4/Swi6)-regulated genes and reveal that binding of Sin3 to SBF-specific promoters is cellcycle-specific regulated. We observe that transcriptional repression of SBFdependent genes in early G1 coincides with the recruitment of Sin3 to specific promoters, whereas binding of Sin3 is abolished from Swi4/Swi6-regulated promoters when transcription is activated at the G1 to S phase transition. Tandem affinity purification uncovered that Sin3 interacts in a complex with the SBF-subunits Swi6 and Swi4. Recruitment of Sin3 to SBF-dependent promoters is largely dependent on Swi6 and Swi4. The timing of Sin3-recruitment to the G1/S-specific promoters is similar to the binding of Stb1 to the same promotors through an interaction with Swi6. Immunopreciptation assays provided evidence that Sin3 interacts with the Start-specific activator Stb1 from late G1 throughout S phase. The observations indicate that recruitment of Sin3 to the G1-Cyclin promoters is not dependent on Stb1. Therefore this work suggests that Stb1 is involved in releasing the promotors from Sin3-dependent repression. We propose a model, in which repression by Sin3/Rpd3 is also abolished by the interaction with Stb1, through the previously described positive Cln1/Cln2-feedbackloop. These results provide evidence, that Sin3 and Rpd3 are directly involved in regulation of G1-Cyclin Transcription (CLN1, CLN2) and therefore are involved in cell size homeostasis at Start. We conclude that the Sin3/Rpd3 histone deacetylase complex helps to prevent premature activation of the S phase in daughter cells.

Abstract

Saccharomyces cerevisiae Zellen kontrollieren ihre Zellgröße in der späten G1-Phase bei Start. In dieser Arbeit wird eine negative Rolle für den Sin3/Rpd3 Histondeacetylasekomplex bei der Regulation der Zellgröße am G1/S-Phaseübergang untersucht. Hefen müssen bis zu einer kritischen Zellgröße wachsen, bevor die Cln3/Cdc28 Proteinkinase in der späten G1-Phase die SBF-abhängige Transkription der G1-Cycline aktiviert und die Zellen in die S-Phase eintreten. In dieser Arbeit wird gezeigt, dass - im Vergleich zum Wildtyp - die G1/Sspezifische Transkription von CLN1, CLN2 und PCL1 in einer sin3 Mutante und einer rpd3 Mutante - bei einer reduzierten Zellgröße - erfolgt. Außerdem führt die Inaktivierung des Transkriptionsregulators SIN3 zu einer teilweisen Suppression des großen Start-Phänotyps von cln3 Mutanten, und bewirkt, dass sin3cln3 Doppelmutanten ihren Zellzyklus bei Wildtypgröße starten. Die Ergebnisse von Chromatin-Immunpräzipitationen zeigen, dass Sin3 und Rpd3 an die Promotoren von SBF (Swi4/Swi6)-regulierten Genen rekrutiert werden und dass die Bindung an SBF-spezifische Promotoren zellzyklusspezifisch reguliert ist. Die Repression der SBF-abhängigen Transkription in der frühen G1-Phase korreliert mit der Rekrutierung von Sin3 und Rpd3 spezifisch an diese Promotoren. Die Dissoziaton von Sin3 von den Swi4/Swi6-abhängigen Promotoren erfolgt, wenn die Transkription am G1/S-Phase Übergang aktiviert wird. Tandem-Affinitäts-Reinigungen zeigten, dass Sin3 mit den SBF-Untereinheiten Swi4 und Swi6 interagiert. Die Rekrutierung von Sin3 an die G1/S-spezifischen Promotoren ist insbesondere abhängig von Swi6 und Swi4. Das Timing der Sin3-Rekrutierung gleicht dem von Stb1 an die selben Promotoren über die Interaktion mit Swi6. Immunpräzipitationen haben gezeigt, dass Sin3 mit dem Startspezifischen Aktivator Stb1, von der späten G1-Phase bis hinein in die S-Phase, assoziiert ist. Die Beobachtungen weisen darauf hin, dass die Rekrutierung von Sin3 an die G1-Cyclin Promotoren nicht von Stb1 abhängt, vielmehr wird vorgeschlagen, dass Stb1 an der Dissoziation von Sin3 vom Promotor beteiligt ist. Die Aufhebung der Sin3/Rpd3-abhängigen Repression erfolgt durch die Phosphorylierung von Stb1 über die kürzlich beschriebene positive Feedback-Schleife. Diese Arbeit beschreibt ein Modell, in dem der Sin3/Rpd3 Komplex an der Zellgrößenhomeostasis bei Start in S. cerevisiae beteiligt ist und eine vorzeitige Aktivierung der S-Phase in Tochterzellen verhindert.
