These secondary metabolites are often indispensable for life of the respective organisms and are produced in an efficient and highly stereo selective way. The efficiency and selectivity of 'nature' is a great challenge for chemists in the laboratory. Although a great variety of methods have been developed to mimic nature, in many cases the chemical synthesis is less stereo selective and/or yields are far below that of the biosynthetic pathway. The development and enhancement of methods is thus a great intention of modern natural chemistry. Beside the total synthesis, the investigation of biosynthetic pathways and the identification of the respective enzymes is a highly interesting field of research. Learning from nature can help to develop new synthetic strategies and methods. Many of the isolated compounds do not only possess the intended biological activity, but have also outstanding effects against pathogens of human diseases. Therefore the isolation and structural elucidation of bioactive compounds as lead structures for new drugs is a challenging and rewarding goal. Despite the great variety and number of natural sources of secondary metabolites, the more and more frequent isolation of already known compounds has become a large problem. To avoid such repeated isolations the methods for the identification and structural elucidation of natural products continuously have been improved. Important contributions are the hyphenation of analytical detectors like MS or NMR with chromatographic facilities e.g. GC, HPLC or UPLC. Combination of liquid chromatography with chiroptical methods like CD spectroscopy sometimes allows the assignment of absolute configurations direct from the extract. The aims of this PhD-thesis were the use of new methods in the synthesis of axially chiral natural products, the elucidation of structures and stereo structures of secondary metabolites and synthetic compounds and the investigation of biosynthetic intermediates.show moreshow less
