Solche Proteine zeichnen sich u.a durch DNA-bindende Eigenschaften aus (Murakami and Okayama, 1995; Allen et al., 1994; Durocher and Jackson, 2002).
  • Kanadaptin is a multidomainprotein, which is expressed in almost all tissue of the rat. It has nuclear import - and export sequences. The import-dependent sequence was identified as NLS1 (189RKRK). The exportsequence might be a leucin-rich domain (414LLQEPELELEAAV). Kanadaptin is also detectable in mitochondrias. In human cells an isoform is produced, which has an aminoterminal insertion containing an FHA. Such proteins show dna-binding features (Murakami and Okayama, 1995; Allen et al., 1994; Durocher and Jackson, 2002). This feature might be the reason for nuclear retention, even after the loss of the integrity of the nuclear core membrane(Hübner et al, 2002). It was also shown, that human kanadaptin cDNA fragment is able to elicite an SOS-Response in an SOS detection E.coli strain(Perkins et al., 1999). These results indicate, that Kanadaptin is involved in DNA and/or DNA-dependent processes. In in-situ-hybridisation Kanadaptin was detetcted mainly in the prox. tubule of the rat kidney. An interaction between Kanadaptin and kAE1 seems less likely, especially since an interaction between the two proteins could not be reproduced in yeast two-hybrid experiments(Wongthida P. et al., 2006). Embryonal kidney cells (HEK293-Cells) also showed no Interaction between the two proteins(Kittanakom et al., 2004). The intracellular function(s) of kanadaptin remain enigmatic and are subject for futher research.show moreshow less
