Zum ersten sollten 4 unterschiedliche hCYR61/CCN1 Promotorkonstrukte, mit den Längen von 200bp-1000bp auf ihre Promotoraktivität getestet werden, um so eine Vorstellung der Regulationsmechanismen des hCYR61/CCN1 Promotors zu gewinnen. Die andere Fragestellung befasste sich mit der Relevanz eines CA-Repeat Polymorphismus innerhalb des hCYR61/CCN1 Promotors, der anhand einer Gesundenpopulation nachgewiesen worden war. Das Interesse war hierbei, ob unterschiedliche CA-Repeat Längen in einem hCYR61/CCN1 Promotorkonstrukt von 450bp Länge auch unterschiedliche Promotoraktivitäten erzeugen. Die hCYR61/CCN1 Promotorkonstrukte der Länge 200bp-1000bp wurden mittels PCR eines Templates (J23219 aus einer Lambda Phagendatenbank) hergestellt. Die Länge und die Sequenz der Promotorkonstrukte überprüften wir durch Sequenzierung und Fragmentanalysen. Ausgewählte hCYR61/CCN1 Promotorkonstrukte wurden daraufhin in ein Luziferase-Vektor-System kloniert. Die entstandenen hCYR61/CCN1 Promotor-Vektorkonstrukte wurden in zwei Zellsyteme, h-fob Zellen (humane Osteoblaten) und T/C28a2 Zellen (humane Chondrozyten) durch Elektroporation transfiziert. Die Zellen wurden über 48h kultiviert, geerntet und die Promotoraktivität in einem Luminometer in Fluoreszenz Units (FU) gemessen. Es wurden zwei verschiedene Kultivierungsbedingungen der beiden Zelllinien geprüft, nämlich eine serumfreie und eine serumhaltige, um einen in der Literatur beschriebenen Seruminduktionseffekt nachweisen und überprüfen zu können. Insgesamt wurden für die hCYR61/CCN1 Promotorkonstrukte der Länge 200bp-1000bp Experimente in h-fob Zellen und T/C28a2 Zellen, jeweils unter serumfreien und serumhaltigen Bedingungen durchgeführt. Für die 450bp langen hCYR61/CCN1 Promotorkonstrukte mit den variierenden CA-Repeats von 17-22 CA-Repeats, wurden ebenso Transfektionsexperimente in beiden Zelllinien und unter beiden Kultivierungsbedingungen durchgeführt. Als Ergebnisse ist zusammengefasst festzuhalten, dass ein statistisch signifikanter Zusammenhang zwischen den hCYR61/CCN1 Promotorkonstrukten unterschiedlicher Länge von 200bp-1000bp und ihrem Promotoraktivitätsniveau besteht. Die hCYR61/CCN1 Promotorkonstrukte der Länge 200bp-800bp besitzen eine sehr hohe basale Promotoraktivität, die um den Faktor 4-5 bei serumfreier Kultivierung über der Hintergrundsaktivität liegt. Die Aktivität des 1000bp Promotorfragmentes ist signifikant niedriger (p=0,024 in h-fob Zellen und p=0,037 für T/C28a2 Zellen bei serumhaltiger wie serumfreier Kultivierung), als die der übrigen Promotorfragmente, die sich untereinander nicht signifikant in ihren Aktivitäten unterscheiden (p=0,57 in h-fob Zellen und p=0,66 für T/C28a2 Zellen bei serumfreire Kultivierung). Es besteht weiterhin ein signifikanter Seruminduktionseffekt, der für alle Konstrukte und beide Zelllinien nachzuweisen ist. Dieser ist bei den hCYR61/CCN1 Promotorkonstrukten der Länge 200bp-1000bp, ebenso wie bei den hCYR61/CCN1 Promotorkonstrukte 450bp mit den variablen 17-22 CA-Repeats nachweisbar (p=0,001 in h-fob Zellen und p=0,002 für T/C28a2 Zellen). Der Seruminduktionseffekt ist für alle Konstrukte proportional und wirkt signifikant stärker auf die T/C28a2 Zellen, als auf die h-fob Zellen. So liegt die durchschnittliche Steigerung der hCYR61/CCN1 Promotoraktivität etwa bei dem Faktor 1,5-3 in h-fob Zellen, in T/C28a2 Zellen jedoch um den Faktor 8-11. Dieser Effekt wirkt auf den Promotor von hCYR61/CCN1, da die basale Hintergrundsaktivität gemessen anhand der Transfektion des leeren Expressionsvektors pgl3-Baisc, davon unbeeinflusst bleibt. Es besteht kein statistisch signifikanter Zusammenhang zwischen den CA-Repeat Polymorphismen 17-22 CA-Repeats in dem hCYR61/CCN1 Promotorkonstrukt der Länge 450bp und deren Promotoraktivitäten (p=0,495 in h-fob Zellen und p=0,514 für T/C28a2 Zellen bei serumfreier Kultivierung und p=0,397 in h-fob Zellen und p=0,488 für T/C28a2 Zellen bei serumhaltiger Kultivierung). Die Konstrukte haben also unabhängig von der Zahl der CA-Repeats keine unterschiedliche Promotoraktivität gemessen im Luziferase Assay. Somit lässt sich in diesem in vitro System kein relevanter Effekt hinsichtlich der funktionellen Relevanz diese CA-Repeat Polymorphismus nachweisen. Weitere Experimente werden Auskunft über kausale Regulationsmechanismen des hCYR61/CCN1 Promotors und die Relevanz von hCYR61/CCN1 innerhalb des Knochenstoffwechsels geben.show moreshow less

The second aim was the characterization of one single CA-repeat polymorphism in the 5´ flanking region of hCYR61/CCN1 in one constant hCYR61/CCN1 promotor fragment. The Hypothesis was that different CA-repeat stretches in a constant promotor fragment cause different levels of promotor activity. Methods: Four different hCYR61/CCN1 promotor stretches from 200bp-1000bp of the 5´ flanking region of hCYR61/CCN1 were produced by PCR from a template of a Phage database. Five different hCYR61/CCN1 fragments with different length of the CA stretch (varying from17-22 CA repeats) in one constant hCYR61/CCN1 promotor fragment length of 450bp, found in a healthy population, were produced by PCR. The control of the PCR products was done by sequencing and by a PCR/fragment analysis protocol. The promotor fragments were cloned into a luciferase assay vector (pgl3-basic) by the TOPO-Blueprint Vector system and transfected into two different cell systems h-fob cells (human Osteoblasts) and T/C28a2 cells (human Chondrocytes) by the use of electroporation. At least 5 different cultivations of each cell system were performed with and without fetal calf serum because the activity of hCYR61/CCN1 was described as growth factor regulated. Measurement of the promotor activity was performed with a luciferase assay system. Results: There is a statistically significant high basic activitity of 3 from 4 promotor stretches of the length 200bp, 450bp and 800bp compared to the base rate (background activity). The biggest promotor fragment 1000bp is significantly lower in promotoractivity (p=0,024 for h-fob cells and p=0,037 for T/C28a2 cells ) compared to the 3 other promotorfragments. This is reproducible for both cell systems and the two types of cell cultivation with and witout fetal calf serum (FCS). There is a significant serum induction effect on the promotor activity of the promotor of hCYR61/CCN1. This is reproducible in both cell systems (p=0,001 for h-fob cells and p=0,002 for T/C28a2 cells). The different length of CA stretches in one constant promotor fragment (450bp) of hCYR61/CCN1 have no statistically significance onto the promotor activity (p=0,495 for h-fob cells and p=0,514 for T/C28a2 cells) Discussion: The significant downregulation of promotor activity in the biggest fragment 1000bp in comparison to the smaller promotor fragments might be caused by a motif called FREAK-1 in 860bp of the 5´ flanking region of hCYR61/CCN1. There is a significant serum induction effect onto the promotoractivity of the promotor of hCYR61/CCN1 despite there is no serum response element onto the promotor like in the murine CYR61 promotor. This fact has to be explained. Further experiments have to explain this interesting effect of downregulation. There seems to exist no relevant functional link between the length of the single CA-stretch in the promotor of hCYR61/CCN1 and the activity of the promotor in vitro.show moreshow less
