Optimierung des In vitro Testverfahrens „Glutathion-Recycling-Assay“zum Nachweis von Schadwirkungen in aquatischen Systemen

Optimization of the in vitro method “glutathione-recycling assay” to proof harmful effects in aquatic systems

  • Es wurden Untersuchungen des „Glutathion-Recycling-Assay“ zur Toxizitätsbewertung von trinkwasserrelevanten Reinsubstanzen und Wasserproben an Jurkat- und HEP G2-Zellen durchgeführt. Dazu wurden die einzusetzende Zellzahl und der Zeitpunkt der Messung optimiert. Des Weiteren wurden die Proteinkonzentration und die eingesäte Zellzahl als Bezugsgröße für die Glutathionmessung untersucht. Zudem wurden zwei Substanzen (TBHP und BSO) auf ihre Eignung zur Positivkontrolle für dieses Verfahren an HEP G2- und Jurkat-Zellen geprüft. Die Validität der Methode wurde anhand statistischer Parameter untersucht.
  • Investigations of the glutathione recycling assay were carried out for the determination of toxicity of pure substances relevant for drinking-water and water samples using Jurkat and HEP G2 cells. Therefore the measuring time and the starting cell number were optimised. Besides the protein concentration and the starting cell number were examined for the use as a reference unit. Furthermore, two substances (TBHP and BSO) were checked for their suitability as positive controls for this method with HEP G2 and Jurkat cells. The validity of the method was examined by calculating of statistical parameters.
