Zusammenfassung
1. Weibliche Wistar-Ratten vom Stamm CFN werden 2 mal tgl. mit 50 MCR mU Calcitonin s.c. behandelt. Nach 12, 24 und 48 Std und 1 und 2 Wochen werden sie getötet und die linke Tibiaepi- und -metaphyse elektronenoptisch untersucht.
2. Die Osteocyten verhalten sich unter Calcitonin zweiphasisch. Sie weisen zwei deutlich differierende formale und Aktivitäts-Bilder auf:
a) Die jungen Osteocyten zeigen wie die Osteoblasten eine Verbreiterung der Mineralisationszone und Formen der Ausschleusung von Ca-Apatit über die Cytoplasmaausläufer. Pathogenetisch könnte dieser Verbreiterung der präparatorischen Verkalkungszone eine Rücknahme der cellulären Hemmzone bis an die Zellmembran zugrunde liegen.
b) Die durch die abnorme Verkalkung resultierende Mikropetrosis veranlaßt die alten Osteocyten zu einer Leistungsumkehr im Sinne einer periosteocytären Osteoclasie. Die Ultrastruktur der alten Osteocyten gleicht derjenigen der Osteoclasten.
3. Es wird zum ersten Mal gezeigt, daß im Mineralisationsvorgang der organischen Matrix Ca-Apatit-Kristalle auch intracellulär, d.h. intraosteocytär und-blastär, ausgefällt werden können.
Summary
1. 4 weeks old female Wistar-rats were treated twice a day with 50 MCR mU of porcine Calcitonin s.c. 12, 24, and 48 hours, and 1 and 2 weeks after starting the investigation the animals were killed and the epi- and metaphysis of the left tibia examined by the electronic microscope.
2. The osteocytes are showing a biphasic behaviour after the treatment with Calcitonin:
a) The young osteocytes showing an enlargment of the zone of demarcation and signs of the output of Ca-apatit-cristals through the polypoids of the cytoplasma.
b) The abnormal mineralisation is leading to micropetrosis and causes the old osteocytes to change their activity. The result is a periosteocytaric osteoclasia. The ultrastructure of the old osteocytes looks like those of osteoclasts.
3. It is shown for the first time, that during mineralisation of the bone matrix the nucleation of Ca-apatit-cristals can start in the cytoplasma of the osteocyts.
