Zusammenfassung: 
Um sich den stetig verändernden Umweltbedingungen anpassen zu können, verfügen Pflanzen, als sessile Organismen, über ein komplexes System aus Abwehrmechanismen. Die Regulation der dabei stattfindenden zellulären Signalwege erfolgt oftmals mithilfe der durch das Ubiquitin-26S Proteasomsystem vermittelten Proteolyse beteiligter regulatorischer Proteinkomponenten. E3 Ubiquitinligasen, u. a. sogenannte Plant U-Box Armadillo Repeat (PUB-ARM)-Proteine, gewährleisten hierbei die Spezifität der Ubiquitinierung durch die Bindung des Substrats.
Im Rahmen dieser Arbeit sollte die Plasmamembran-assoziierte PUB-ARM E3 Ubiquitinligase SAUL1 charakterisiert werden. Vorangegangene Studien haben gezeigt, dass SAUL1 als Suppressor bei temperaturbedingtem Zelltod und Seneszenz fungiert. Ferner scheint SAUL1 als positiver Regulator an der Immunantwort in A. thaliana beteiligt zu sein. In einem ersten Projekt wurde mithilfe von Genexpressionstudien der Einfluss von SAUL1 und den nächstparalogen PUB-ARM Proteinen PUB43 und PUB42 auf die Expression Immunabwehr-spezifischer Gene analysiert. Infolge der Überexpression von SAUL1 und PUB43 konnte ein deutlicher Anstieg der PR1- und PR2-Genexpression sowie ein Autoimmunitätsphänotyp festgestellt werden. Ein Zusammenhang der beiden Proteine bei der pflanzlichen Immunantwort kann daher vermutet werden. Im Gegensatz dazu ergab ein Vergleich der PR-Genexpression von pub42-2 saul1-1 Doppelmutanten mit saul1-1 Mutanten keinen additiven Effekt des Ausschaltens von PUB42 auf die Genexpression. Zusätzlich konnte kein temperatursensitiver Phänotyp in den pub42-2 Mutantenlinien ermittelt werden, sodass PUB42 keinen Einfluss auf die Genexpression bei der Immunantwort in A. thaliana zu haben scheint.
Im Fokus dieser Arbeit lag die Identifizierung von SAUL1-Interaktionspartnern, um mögliche Substrate der E3 Ubiquitinligase zu finden. Hierbei wurden zwei verschiedene Strategien verfolgt. Zum einen konnten mithilfe einer Tandemaffinitätsaufreinigung mehrere putative Interaktoren von SAUL1 aus pflanzlichen Zellkulturen isoliert werden. Hierbei wurden überwiegend Proteine identifiziert, die im Zellkern lokalisiert sind. Die Assozation von SAUL1 mit dem Kinesin-5-Protein AtKRP125b konnte mithilfe der bimolekularen Fluoreszenzkomplementation (BiFC) in planta bestätigt werden. Darüberhinaus wurde die subzelluläre Lokalisierung von AtKRP125b im Zellkern von A. thaliana nachgewiesen. In einem zweiten Ansatz wurden BiFC-Interaktionsstudien mit Proteinen, die ähnlich wie SAUL1, bei der pflanzlichen Immunantwort beteiligt und an der Plasmamembran lokalisiert sind, durchgeführt. Hierbei konnte eine Interaktion von SAUL1 mit dem Copine Protein BON1, nicht jedoch mit dem BON1-Interaktionspartner BAP1 und der Immunrezeptorkinase LORE ermittelt werden. In Lokalisierungsstudien konnte gezeigt werden, dass die intrazelluläre Lokalisierung von SAUL1 und BON1 an der Plasmamembran unabhängig vom Interaktionspartner erfolgt. Darüberhinaus wurde eine Wechselwirkung zwischen PUB43 und den beiden Proteinen BON1 und BAP1 beobachtet, sodass eine Komplexbildung vermutet werden kann.
In einem weiteren Projekt wurde die Topologie und Assoziation von SAUL1 an der Plasmamembran untersucht. Anhand der direkten stochastischen optischen Rekonstruktionsmikroskopie konnten Hinweise auf oligomere Strukturen der E3 Ubiquitinligase erhalten werden, die die durch vorangegangene Studien vermutete Tendenz von SAUL1 zur Bildung von höher geordneten Strukturen, untermauern. Des Weiteren konnte durch eine selektive Behandlung und Analyse der Gesamtmembranfraktion festgestellt werden, dass SAUL1 als peripheres Membranprotein vermutlich über hydrophobe Wechselwirkungen mit der Plasmamembran in A. thaliana assoziiert ist.

As sessile organisms plants possess a complex system of defence mechanisms in order to adapt to changing environmental conditions. The regulation of the associated cellular signaling pathways is achieved by the ubiquitin-26S proteasome system resulting in proteolysis of participating regulatory protein components. E3 ubiquitin ligases, e.g. Plant U-Box Armadillo Repeat (PUB-ARM)-proteins ensure the specificity of the ubiquitination process.
The aim of this work was the characterization of the plasma membrane-associated PUB-ARM E3 ubiquitin ligase SAUL1. Previous studies showed that SAUL1 functions as suppressor of temperature-affected cell death and senescence. Furthermore SAUL1 seems to be involved in the immune response in A. thaliana as a positive regulator of PTI. In the first project, gene expression studies of immune response-associated genes were performed in order to analyze the role of SAUL1 and its closest paralogous proteins PUB43 and PUB42. As a result of SAUL1 and PUB43 overexpression the expression level of PR1 and PR2 increased and plants exhibited phenotypic symptoms of autoimmunity, indicating a function of the proteins in plant immune responses. In contrast, the comparison of PR-gene expression in pub42-2 saul1-1 double mutant and saul1-1 mutant plants exhibited no additive effect of the PUB42-knock-out on gene expression. In addition, pub42-2 mutants did not show the typical saul1-1 temperature-sensitive phenotype. Therefore PUB42 does not seem to affect gene expression during immune response in A. thaliana.
The main topic of this work was the identification of SAUL1 interaction partners in order to find potential substrates of the E3 ubiquitin ligase. Therefore two different strategies were followed. On the one hand, various putative SAUL1 interactors were identified using tandem affinity purification. The majority of identified proteins are localized in the nucleus. By means of bimolecular fluorescence complementation (BiFC), the in planta association of SAUL1 with the kinesin-5 protein AtKRP125b was confirmed. Furthermore, the subcellular localization of AtKRP125b to the nucleus was proven. On the other hand, presumed interactions of SAUL1 with proteins, which are involved in the immune response and localized to the plasma membrane, were analyzed via BiFC. SAUL1 interacts with the copine protein BON1, but neither with its interaction partner BAP1 nor with the immune receptor kinase LORE. Localization studies revealed that SAUL1 and BON1 associate to the plasma membrane in an interaction partner-independent manner. Moreover PUB43 was shown to interact with BON1 and BAP1. Based on this result a complex formation between these three proteins is presumed.
In another project, the topology and association of SAUL at the plasma membrane was analyzed. The direct stochastical optical reconstruction microscopy revealed indications of oligomer structures of the E3 ubiquitin ligase, substantiating previous observations presuming the tendency of SAUL1 to build higher organized structures. Furthermore the selective treatment and analysis of the total membrane fraction exhibited that SAUL1 as a peripheral membrane protein is probably associated with the plasma membrane via hydrophobic interactions.
