Abstract: Im Rahmen dieser Arbeit wurde der Abbauweg des neuartigen Zuckers 1,5-Anhydro- D-fructose (1,5-AF) in Sinorhizobium meliloti 1021, einem Modellbakterium dessen Genomsequenz bekannt ist, aufgeklärt. Die beteiligten Enzyme Anhydrofructose- Reduktase (AFR) und eine P450-Monooxygenase (CYP) wurden in Zellextrakten nachgewiesen und charakterisiert. Die postulierten Reaktionen wurden mit den rekombinanten Enzymen in vitro verifiziert. Der erste Reaktionsschritt wird durch AFR katalysiert, die 1,5-AF stereoselektiv zu 1,5-Anhydro-D-mannitol (1,5-AM) reduziert. Die AFR aus S. meliloti 1021 ist ein dimeres, strikt NADPH-abhängiges Enzym 74,1 kDa und hat 80 % Sequenzidentität zu AFR aus S. morelense S-30.7.5. Die für S. morelense S-30.7.5 postulierte Hydroxylierung von 1,5-AM an C-1 zu D- Mannose durch ein CYP-System konnte experimentell bestätigt werden. Zwei putativ für CYP kodierende ORFs wurden aus S. meliloti kloniert und charakterisiert. Nur die aus dem ORF Y01812 erhaltene Monooxygenase konnte in aktiver Form exprimiert werden. Für den Elektronentransfer von NADH auf die MO1812 wurden aus P. putida die Gene der Putidaredoxin-Reduktase (camA) und des Putidaredoxins (camB) kloniert und mit der MO1812-(His)6 fusioniert. Im Test mit 1,5-AM war das Fusionsprotein aktiv. Die Beteiligung der Monooxygenase an dem Abbauweg von 1,5-AF in S. meliloti 1021 wurde zusätzlich durch Deletion des Gens Y01812 verifiziert, wobei die Fähigkeit des Bakteriums zum Wachstum auf 1,5-AF verloren ging.
A catabolic pathway of the new sugar 1,5-anhydro-D-fructose (AF) in Rhizobia was clarified in the frame of this work. S. meliloti 1021 strain was chosen as a model due to the complete genetic data available. The enzymes of this pathway were identified in cellextracts and characterized. Their participation in these reactions was verified. The first reaction is catalysed througt AF reductase which reduces stereoselective AF to 1,5-anhydro-D-mannitol (AM). The afr-gene from S. meliloti has 1002 bp and the derived polypeptide showed 80 % identity with the AFR from S. morelense. The AF reductase from S. meliloti is a strictly NADPH dependent dimeric protein of 74 kDa and it has 80 % homology to the AFR from S. morelense S-30.7.5. The for S. morlense S-30.7.5 C-1 postulated oxygenation of 1,5-AM to the D-mannose through a CYP system could be verified. Two putative CYP coding ORF';s from S. meliloti were cloned and characterized. Only the monooxygenase from the ORF Y01812 could be expressen in an active form. For the electron transport from NADH the MO1812 two genes: putidaredoxin reduktase (camA) and putidaredoxin (camB) from P. putida were cloned and fused with the monooxygenase MO1812-(His)6. The fusion protein was in the test with 1,5-AM activ. To confirm the involvement of this monooxygenase in the 1,5-anhydro-D-fructose pathway the monooxygenase-gene was accessorily knocked out in S. meliloti 1021, what resulted in no-growth of the organism on 1,5-AF.
