Kurzfassung
Ein zentraler Aspekt im Tissue Engineering ist das Scaffold. Durch seine Beschaffenheit und den direkten Kontakt zu den Zellen beeinflusst es nachhaltig die Entwicklung des tissue-engineerten Konstruktes. Besonders attraktiv sind autologe Materialien, wie Fibrin. Zur Optimierung von autologen PFP/Fibrin-Gel-Scaffolds wurde in dieser Arbeit Plättchenfreies Plasma (PFP), Plättchenarmes Plasma (PPP) und Plättchenreiches Plasma (PRP) als Ausgangsmaterial für die autologe Scaffoldproduktion verglichen. Es wurden drei verschieden stark konzentrierte PPP- bzw PRP-Gele verwendet. Dazu wurden Untersuchungen zur Mikrostruktur der Gele, zur mechanischen Stabilität, Proliferation, EZM-Bildung und zur Gewebeneogenese vorgenommen. Außerdem wurde der Einfluss der Wachstumsfaktoren bFGF und TGF-ß auf Myofibroblasten/HUASMCs untersucht.Zusammenfassend zeigen die Ergebnisse, dass die hoch konzentrierten Plasma-Gele im Gegensatz zu den PFP/Fibrin-Gelen bessere Ergebnisse in allen Versuchen erzielt haben. Vor allem konnte gezeigt werden, dass auch ohne die Zugabe von FCS die Zellen, die in PPP- und PRP-Gelen kultiviert wurden proliferieren, Kollagen bilden und die Migration von Zellen positiv beeinflussen. Da zwischen den PPP- und PRP-Gelen kaum Unterschiede festgestellt wurden, sondern nur im Vergleich zu PFP/Fibrin-Gelen, scheint der entscheidende Faktor das Resuspendieren der Fibrinogen-Pellets nach der Kryopräzipitation in Plasma zu sein. Die Ergebnisse legen nahe, dass in nachfolgenden Studien PRP oder zumindest Plasma als Resuspensionsmedium nach der Kryopräzipitation genutzt bzw. auf Waschschritte verzichtet werden sollte, um ein verbessertes Ausgangsmaterial für fibrinbasierte Scaffolds nutzen zu können. Weiterhin sollte untersucht werden, wie die mechanische Stabilität zu optimieren ist und die Ausgangsblutmenge reduziert werden kann.Myofibroblasten/HUASMCS haben keine Rezeptoren für TGF-ß, jedoch für bFGF. Es konnte kein Effekt auf die mechanische Stabilität, die Kollagensynthese, Proliferation, Apoptose oder Nekrose von Myofibroblasten/HUASMCs durch bFGF festgestellt werden. Es ist sogar anzunehmen, dass bFGF die Kollagensynthese negativ beeinflusst. In nachfolgenden Studien sollten weitere Konzentrationen sowie Kombinationen von verschiedenen Wachstumsfaktoren auf ihre Wirkung auf HUASMCs untersucht werden, da diese Faktoren sich auch gegenseitig beeinflussen und eventuell erst durch die richtige Kombination ihre gewünschte Wirkung auf die Zellen zeigen.

One central aspect in tissue engineering is the scaffold. Due to its properties and the direct contact to cells, it strongly influences the development of the tissue engineered construct. Especially attractive are autologous materials, like fibrin. To optimize autologous PFP- and fibrin-scaffolds this work compared the basic materials platelet free plasma (PFP), platelet poor plasma (PPP), and platelet rich plasma (PRP) as autologous materials for scaffold production. Three differently concentrated PPP and PRP-gels were used. Therefore the microstructure of gels, the mechanical strength, proliferation, ECM-development and neogenesis were analyzed. Furthermore the influence of the growth factors bFGF and TGFß on myofibroblasts/HUASMCs was investigated.In conclusion all experiments show better results for the highly concentrated plasma-gels in contrast to the PFP/fibrin-gels. In particular, it could be shown that cells cultivated in PPP- and PRP-gels proliferated, produced collagen and migration was positively influenced also without the addition of FCS. Between PPP and PRP the differences were minor, just in comparison to PFP/fibrin-gels the difference were strong. So it seems that the key element is the resuspention of the fibrinogen pellet after cryoprecipitation in plasma. The results suggest that in follow-up studies the resuspention medium should be plasma and washing steps should be avoided to obtain a better basic material for fibrin based scaffolds. Furthermore the optimization mechanical strength and the reduction of blood volume should be investigated in upcoming studies.Myofibroblasts/HUASMCS had no receptors for TGF-ß, but for bFGF. Still no effect on mechanical strength, collagen synthesis, apoptosis, proliferation and necrosis on myofibroblasten/HUASMCs could be observed due to bFGF. More likely it is to expect that bFGF negatively influences collagen synthesis. In follow-up studies different combinations of growth factors should be investigated, because it is likely that those factors influence each other. The investigation of different combinations of growth factors could lead to the right combination for the desired effects on HUASMCs.
