Inhibitoren für Cysteinproteasen : Synthese, Screening und Methodenentwicklung

Basierend auf der Bedeutung der Proteasen für das (Über-)Leben eines Organismus stehen Enzyme seit einigen Jahren im Fokus der Wirkstoffforschung. Ziel hierbei ist es, biologisch relevante Signalwege durch spezifische Hemmstoffe (Inhibitoren) zu blockieren. Dadurch kann zum Beispiel die Replikation eines Virus oder die Ausbreitung eines Tumors verhindert werden. Es hat sich gezeigt, dass vor allem Cysteinproteasen ein besonders attraktives Ziel für die Wirkstoffentwicklung sind. Ihre fundamentale Rolle im Entwicklungsprozess von viralen parasitären Infektionen haben sie zu interessanten Angriffspunkten für die Erforschung neuer Therapeutika gemacht. Ziel dieser Arbeit war es, durch die Anwendung unterschiedlicher Methoden und Strategien, Inhibitoren für verschiedene Cysteinproteasen zu entwickeln. Die Herangehensweise an die Problemstellungen erfolgte durch die Anwendung von geeigneten Synthesetechniken, Screeningbedingungen und Analysemethoden. Untergliedert wurde die Arbeit in drei Abschnitte, die sich jeweils mit einer gezielten Fragestellung beschäftigten. <br> Im ersten Teil dieser Arbeit war geplant, die in einem früheren Projekt entdeckten Cysteinproteaseinhibitoren in ihren Hemmeigenschaften noch zu verbessern und Informationen über den Zusammenhang zwischen der Wirkung sowohl am Erreger als auch an den isolierten Zielenzymen und der Struktur der Inhibitoren zu erhalten. Für diese Verbindungsklasse waren P. falciparum (Malaria) und das zugehörige Enzym Falcipain-2, sowie T. b. brucei (Nagana) und das aus T. b. rhodesiense (Afrikanische Trypanosomiasis) stammende Enzym Rhodesain als biologische Angriffspunkte identifiziert worden. <br> Im zweiten Teil stand die Entwicklung einer Screeningmethode im Vordergrund, welche es in Zukunft erlauben soll, die Hauptprotease des HCoV 229E mit Hilfe von harzgebundenen Verbindungen in einem „on bead“ Screening zu untersuchen. Ein Ziel dieser Arbeit war es, durch die Wahl geeigneter Assaybedingungen und harzgebundener Verbindungen eine Screeningmethode an der viralen Cysteinprotease HCoV 229E MPro zu entwickeln. Das Zielenzym stellt einen vielversprechenden Angriffspunkt für die Entwicklung von Inhibitoren dar. Durch die enge strukturelle und evolutionäre Verwandtschaft mit der Hauptprotease des SARS CoV können die Ergebnisse auch für die Entwicklung von Inhibitoren an dieser Protease genutzt werden. Dafür ist die Erforschung von Leitstrukturen notwendig, um geeignete Strukturen zu ermitteln, die Hemmeigenschaften an der Protease aufweisen.<br> Ziel des dritten Projekts war die Identifizierung von Inhibitorstrukturen mit Hilfe einer dynamischen kombinatorischen Bibliothek. Verschiedene Bausteine wurden dazu durch eine reversible Reaktion ins Gleichgewicht gebracht und durch die Zugabe eines Templats – im vorliegenden Fall vorwiegend Cysteinproteasen – die Bildung von thermodynamisch begünstigten Verbindungen verstärkt. Auf diese Weise lassen sich potentielle Inhibitorstrukturen finden. Die zentrale Herausforderung in diesem Projekt bestand sowohl in der Auswahl der Bibliothekszusammensetzung, der Synthese der Komponenten, als auch der Durchführung und Anpassung der Screeningmethode an die Assaybedingungen.

Based on the importance of proteases in the lifecycle of an organism, enzymes are in the focus of drug discovery for several years. The aim in this case is to block biologically relevant pathways by specific inhibitors. Thus for example, the replication of a virus or the spread of a tumor can be prevented. It has been found that especially cysteine proteases are a particularly attractive target for drug development. Their fundamental role in the development process of viral and parasitic diseases have made them an attractive target for the research of new drugs.<br> The aim of this study was to develop inhibitors for various cysteine proteases with the use of different methods and strategies. The approach includes the application of appropriate synthesis techniques, screening conditions and analytical methods. Subdivided into three sections, each part of the work focused on different questions or tasks. The plan for the first part of this work was to optimize cysteine protease inhibitors known from a former project and get information about the relationship between the effect at the pathogen respectively at the isolated enzyme and the structure of the inhibitor. As biological targets for this compound class P. falciparum (Malaria) and the corresponding enzyme falcipain-2, as well as T. b. brucei (Nagana) and the enzyme rhodesain from T. b. rhodesiense (African Trypanosomiasis) were identified.<br> The development of a screening method for the mainprotease of HCoV 229E was in the spotlight of the second part. One goal of this study was to develop a screening method for the viral cysteine protease HCoV 229E MPro with the choice of appropriate assay conditions and resin-bound compounds. This enzyme represents a promising target for the design and development of inhibitors. Due to the close structural and evolutionary relationship with the mainprotease of SARS CoV, the results from HCoV 229E MPro can be used for the SARS protease to create inhibitors. For the exploration of lead structures it is necessary to determine appropriate structures that show inhibitory properties at the protease. The aim of the third project was the identification of inhibitors with the help of a dynamic combinatorial library. Several molecules were brought to equilibrium by a reversible reaction and the addition of a template, which enhances the formation of thermodynamically favored compounds. In this case mainly cysteine proteases were used. In this way potential inhibitor structures can be found. The main challenge in this project included the development of the library composition, the synthesis of the components, as well as the implementation and adaptation of the screening method to the assay conditions.
