Analyse von genetischen Läsionen in B-Zell-Non-Hodgkin-Lymphomen und im klassischen Hodgkin-Lymphom.

In dieser Arbeit konnten neue Erkenntnisse über die Vielfalt genetischer Läsionen und über die unterschiedlichen Mechanismen der deregulierten NF-kB-Ausprägung in den verschiedenen B-NHLs und dem cHL gewonnen werden. Durch die Abwesenheit inaktivierender Mutationen im TNFAIP3-Gen konnte gezeigt werden, dass dieses Gen bei der konstitutiven Ausprägung des NF-kB-Signalweges im Gegensatz zu vielen verschiedenen anderen Lymphomen in CLL-Zellen keine Rolle spielt. Die Vermutung, dass TNFAIP3 das Zielgen der selteneren 6q Deletion in einigen CLL-Fällen ist, wurde also widerlegt. Der Befund, dass TRAF3 nur in einer cHL-Zelllinie durch eine Deletion inaktiviert wurde und in allen weiteren Zelllinien und in den sieben Primärfällen nicht von inaktivierenden Mutationen oder Deletionen betroffen ist, demonstriert, dass die TRAF3-Inaktivierung in der Regel nicht zur konstitutiven Ausprägung des NF-kB-Signalwegs in HRS-Zellen beiträgt, sondern eher ein seltenes Ereignis darstellt. Weiterhin wurde mit zwei in dieser Arbeit etablierten MYC-LDI-PCR-Ansätzen in einem von 13 Fällen das bislang in MYC-Translokationen unbeschriebene Partnertgen SOCS1 identifiziert. Da SOCS1 ein bekannter Tumorsuppressor ist, ist die Fusion des Onkogens MYC mit einem Tumorsuppressor besonders interessant. Der Mechanismus, der aufgrund dieser Translokation auf die beiden Gene wirkt, muss aber noch geklärt werden. In dieser Arbeit konnten in sieben von 28 B-NHL-Fällen mit Hilfe zum Teil neu etablierter LDI-PCR-Ansätze IgH-assoziierte Translokationen amplifiziert und sequenziert werden. Vier dieser Translokationen stellten sich als bislang noch völlig unbeschriebene Partner einer solchen Translokation in DLBCL heraus. In der überwiegenden Mehrheit dieser Translokationen wurde die kodierende Sequenz zerstört. Es bleibt noch zu überprüfen, ob dadurch ein bislang noch unbekannter Tumorsuppressor inaktiviert wird oder ein entfernt liegendes bis jetzt noch nicht identifiziertes Onkogen dereguliert wird. In einem Fall konnte allerdings das IRF4-Gen als beteiligtes Onkogen erkannt werden, was zur Charakterisierung einer neuen Untergruppe des GCB-DLBCL führte. Die hier durchgeführten Untersuchungen zeigen also nicht nur, dass DLBCL ein weitaus heterogeneres Lymphom ist, als bislang angenommen, sondern bieten mit der Identifizierung des PVRL2-Gens auch ein weiteres Beispiel dafür, dass in B- und T-Zell-Lymphomen die gleichen Bruchpunkte in Translokationen betroffen sein können.

In this Ph. D. Thesis it was possible to get new insight about the variety of genomic lesions in B-NHLs and gain new knowledge about the mechanisms of deregulated NF-kB signaling in B-NHLs and cHL. The absence of inactivating mutations in TNFAIP3, indicates that, in contrast to other lymphomas, this gene was not involved in the constitutive activation of the NF-kB pathway in CLL. The supposition that TNFAIP3 is the target of the rare 6q deletion in some CLL-cases was therefore disproved. TRAF3 was inactivated by a deletion only once in a cHL cell line, but it was not affected by inactivating mutations or deletions in all other analyzed cell lines and in seven primary cHL-cases. This demonstrates that TRAF3 inactivation does not account normally for the constitutive NF-kB-expression in HRS-cells, but seems to be an uncommon event. In this work, two MYC-LDI-PCR approaches were established and used to amplify in one out of 13 B-NHL-cases a translocation with SOCS1 as a to date unknown translocation partner of MYC-translocations. Because SOCS1 is a known tumor suppressor, a translocation juxtaposing SOCS1 to the MYC-oncogene is of great interest. The mechanism affecting both translocated genes remains to be identified. The partly new established igH-LDI-PCR approaches in this work were successfully used to amplify and sequence the translocation partner of seven out of 28 DLBCLs with IgH-associated translocations. In four of these cases the translocation partners were so far not described in translocations involving the IgH-locus in DLBCLs. In the majority of the cases the translocation disrupted the coding sequence of the partner gene. It remains to clarify, if the translocations inactivate an unknown tumor suppressor or deregulate a distant unknown oncogene. In one of the cases IRF4 was identified as target oncogene of the translocation; this finding led to the characterization of a novel subtype of GCB-DLBCL. The results presented here show that DLBCL is more heterogeneous as previously expected and the identification of PVRL2 as translocation partner shows another example of translocations affecting the same breakpoint in B- and T-cell lymphomas.
