Abstract

Im Rahmen dieser Arbeit wurden die chemischen und strukturellen Merkmale sowie die biologische Bedeutung des Schreckstoffs des Danio-Fisches Malabarbärbling untersucht. Die Isolierung und Identifizierung der Pheromon-Substanz wurde mit Hilfe der Methoden (HPLC, GFC, UV-VIS, NMR and LDIMS) durchgeführt. 3 mg des isolierten natürlichen Pheromons wurden verwendet, um dessen Struktur mit instrumentellen analytischen Methoden zu identifizieren. Abschließend erfolgte der Vergleich der untersuchten Daten des Schreckstoffs mit denen der synthetischen Komponente. Das natürliche Alarmpheromon wurde als 7-Hydroxybiopterin identifiziert. Bei Biotests mit 5 mL einer 0,4 myM 7-Hydroxybiopterin-Lösung in 20 L Aquarien konnte eine Schreckreaktion ausgelöst werden. Um die Spezifität und Selektivität des Pheromons zu verifizieren, wurde der Biotest mit verschiedenen Substanzen aus der Verbindungsklasse der Pterine und Purine durchgeführt. Wie die Ergebnisse des Biotests zeigen, handelt es sich bei 7-Hydroxybiopterin um einen Schreckstoff mit einer hochspezifischen Pheromonstruktur. Kleine Änderungen der Struktur führten zum Verlust der pheromonalen Aktivität.

Within the scope of this work, the chemical structural features and biological signifiance of the alarm pheromone of giant danio Danio malabaricus were investigated. The isolation and identification of the pheromone substance was carried out by deploying the methods (HPLC, GFC, UV-Visible, NMR and LDIMS). Each step of the separation procedure was accompanied by bioassays with Danio malabaricus. An amount of 3 mg isolated natural pheromone was obtained to identify its structure by using instrumental analytical methods. The investigated data of active substance compared with those of proposed synthetic compounds. Consequently the natural alarm pheromone, 7-hydroxybiopterin is elucidated. The lowest bioactive amount, that induced a fright reaction in bioassays, was 5 mL of 0,4 my-M 7-hydroxybiopterin solution in 20 L aquarium. For specificity and selectivity of the pheromone the bioassays were investigated with several substances, which are structure related with pterins and purins. The results presented here suggested that 7-hydroxybiopterin is a highly specific pheromone structure. Little changes of chemical structure of this 7-hydroxybiopterin led to lose of pheromonal activity.
