Abstract

Oxidativer Stress ist an der Pathogenese von neurodegenerativen Erkrankungen beteiligt. Oligodendrozyten (ODC) sind die myelin-bildenden Zellen des ZNS und reagieren auf oxidativen Stress besonders empfindlich. Vor kurzem konnte gezeigt werden, dass neben der Bcl-2-Familie vor allem die Mitochondrien eine wichtige Rolle in der Koordinierung des programmierten Zelltodes (PCD) spielt. Um die molekularen Mechanismen zu überprüfen, die dem H2O2-vermittelten PCD in ODC zugrundeliegen, wurden die mitochondriale Funktion und Signaltransduktionswege untersucht. Die Experimente zeigten eine frühe Freisetzung von Cytochrom C aus den Mitochondrien in das Cytosol und in einen Zusammenbruch des mitochondrialen Membranepotentials (Psi m) nach Behandlung mit H2O2. Die Behandlung ergab ferner eine Aktivierung der Caspase-9 und Caspase-3. Oxidativer Stress führt weiter zu einer Aktivierung von ERK-1,2, einer Kinase die an der Regulierung des Zelltodes und Überlebens beteiligt ist. Die Daten zeigen, dass der mitochondriale Weg am durch oxidativen Stress ausgelösten PCD in ODC beteiligt ist.

Oxidative stress has been implicated as a causal factor of neurodegenerative diseases. Oligodendrocytes (ODC) are the myelin forming cells of central nervous system and are especially vulnerable to oxidative stress. Recently, it was found that besides the Bcl-2 family, mitochondria play a pivotal role in coordinating programmed cell death (PCD). To examine the molecular mechanisms underlying H202-mediated PCD in ODC, we have investigated mitochondrial function and integrity and signal transduction pathways. Time course experiments showed an early release of cytochrome c from mitochondria into the cytosol and a slight reduction of the mitochondrial membrane potential (psi m) after H2O2 exposure. Concomitantly, the treatment with H2O2 resulted in caspase-9 and caspase-3 activation as demonstrated by Western Blot. Additionally, oxidative stress caused the activation of ERK 1,2, a kinase participated in the regulation of cell death and survival. The data suggest that the mitochondrial pathway is involved in PCD in ODC after oxidative stress.
