Abstract

Im Rahmen dieser Arbeit wurden erste Grundlagen für die Erzielung hormetischer Effekte in der Haut über eine Inhibition des proteasomalen Apparates geschaffen. Hierfür wurden humane Fibroblasten und Keratinocyten mit MG132, einem synthetischen Inhibitor des Ubiquitin-Proteasomen-Systems, behandelt und folgende Parameter charakterisiert: (1) Die Induktion von Hitzeschockproteinen (Hsps), (2) die Aktivierung von Makroautophagie (MA), (3) posttranslationale Veränderungen von Proteinen durch SUMO (small ubiquitin related modifier). Die Ergebnisse zeigen, dass (1) Keratinocyten empfindlicher auf proteasomalen Stress reagieren als Fibroblasten, (2) MG132 in beiden Zelltypen zu einer Induktion von Hsps und MA führt, (3) die MG132-induzierte MA in beiden Zelltypen ein protektiver Mechanismus ist, (4) nach proteasomaler Inhibition ubiquitinierte und SUMOylierte Proteine akkumulieren und unlöslich werden. SUMO-1 könnte neben Ubiquitin als sekundäres Signal für den proteasomalen Abbau dienen.

In the course of this thesis basic ways that ensure the achievement of hormetic effect in the skin are given by inhibition of the proteasomal system. For this purpose human fibroblasts and keratinocytes were treated with MG132, a proteasomal inhibitor, and the following parameters were characterized: (1) Induction of heat shock proteins, (2) induction of macroautophagy (MA), (3) posttranslational modifications of proteins by SUMO (small ubiquitin related modifier). The results show that (1) keratinocytes are much more sensitive to proteasomal stress than fibroblasts, (2) MG132 leads to an induction of Hsps and MA, (3) MG132-induced MA functions as a protective mechanism, (4) ubiquitinated and SUMOylated proteins accumulate and get insoluble. SUMO-1 could act as an second signal for proteasomal degradation beneath ubiquitin.
